A Correlation Study On Pathogenesis In Th17,Treg Cells And Related Cytokines In Pemphigus

Purpose:Pemphigus is a group of autoimmune bullous diseases characterized by a loss of cell adhesion and blister formation within the epidermis in the skin.The immunological basis for pemphigus is well established and includes auto-antibodies targeted against two proteins in the cell adhesion structure:Desmogleinl and Desmoglein3.Many of the B-cell behavior is regulated by the T cells.Its pathogenesis is complicated.The CD4+T lymphocyte is categorized into Th1,Th2and Treg cell.Thl7cell,a kind of CD4+T cell subpopulation is found recently. Th17cell has independent differentiate and growth regulatory mechanism. The function and differentiation of Th17and Treg cell against one another,which plays an important role in autoimmune diseases. It is rare reported on the relate between Th17,Treg cell and IL-17,TGF-β with the pathogenesis of pemphigus. In order to find this answer, we examine the expression of Th17and Treg cell in peripheral blood PBMC by FCM and related factors IL-17,TGF-β in serum by ELISA in patient,and compared to normal person,in order to study the role of Th17,Treg cell and IL-17,TGF-β in Pemphigus.Methods:Samples come from The Second Affiliated Hospital of Kunming Medical University. The pemphigus presents are39, the infected patients are20, the uninfected patients are13, the active patients are6, the remission patients are6(The return of active patients after one month remission) and the control are30.We use the flow cytometry to test the Th17and Treg cell and ELISA to test IL-17and TGF-β in six groups above mentioned.Use the SPSS17.0to analysis the results.Results:1. The Th17in the pemphigus patients are higher than control group.2.The Treg cells in the pemphigus patients are higher than control group, The Treg cells in the infected group and higher uninfected group.3. The serum level of IL-17in pemphigus present are higher than control group.4. The serum level of TGF-P in pemphigus present are higher than control group.Conclusion:The abnormal expression of Thl7/Treg cells in the pemphigus; The serum level of IL-17, TGF-P in the pemphigus patients are higher.Th17and Treg cells that participate in the pathogenesis of the vascular diseases.
