Objective:1. Research the survival effect on the human lung adenocarcinoma cell A549cells line after various concentration Genistein treated.2. To study the expression of Bax、Bid、caspase-3、caspase-8and caspase-9after various concentration Genistein treated.Methods:1. To measure the viability of human lung adenocarcinoma A549cells that variance levels of genistein treated between24,48and72hours by MTT assay. Calculated drug median inhibitory concentration(IC50).2.Westren blotting analysis the expression changes of Bax, Bid, caspase-3, caspase-8and caspase-9protein after different concentration genistein effect lung adenocarcinoma A549cellsResults:1, Genistein significantly inhibited the proliferation of human lung adenocarcinoma cell line A549and the obvious proliferation inhibition effect of A549cells was concentration-dependent and time-dependent manner. The median inhibitory concentration (IC50) is45.9mmol/1.2. With the β-actin as the internal reference control, the western blotting show that:after different concentrations of Genistein treated human lung adenocarcinoma A549cells, the expression of Bax and Bid were enhanced; the expression of caspase-3, caspase-8and caspase-9were increased;Western boltting revealed that in depends on concentration-dependent changes.Conclusion:1.The effect of genistein in promoting the apoptosis of lung adenocarcinoma A549cells in a concentration-dependent and time-dependent manner.2. Genistein up-regulated the expression of Pro-apoptotic proteins Bax and Bid to induce apoptosis of human lung adenocarcinoma A549cells.3. Genistein may induces the apoptosis of human lung adenocarcinoma A549cells by caspase-8/caspase-3cascade and caspase-9/caspase-3cascade apoptosis key pathways.
