Glioma, a disease of the nervous system, it is difficult to cure, the mortality rate is up tothirty thousand people in China, and is ranked the second of children malignant tumor, mainreasons of the disease is physical and chemical factors and genetic factors, against glioma, themain method is surgical excision, but the disease still has the poor prognosis, and it is easy torelapse, this is a dangerous drawback, so we would like to search for the treatment of glioma,first of all, starting from the basic research, finding out the tumor related genes, excavatingthe development mechanisms of the tumorigenesis, eventually found the tumor therapeutictargets, clinically for tumor treatment.In recent years, the study of finding the interaction of Netrin-1and its receptor, incontrol tumor occurrence and development. Netrin-1act as live factor through the receptorUNC5H and DCC. So we aimed to explore the expression of Netrin-1in different levels ofhuman glioma, and Netrin-1and the receptor Neogenin interaction law and expression, thento discuss the influence of the Netrin-1and the mechanism of tumor development.The experiment by immunohistochemical testing the expression of Netrin-1and itsreceptor Neogenin in different levels of glioma. The laboratory work has shown theexpression of Neogenin, with the grade of tumor changes,reducing in the glioma. Throughthe PCR and Western blot analysis here, we found that as tumor level increased, theexpression of Netrin-1increase, at the same time, the expression of the Neogenin is oppositerelationship.The experiments show that, with the increased level of tumor, malignant degree ofincrease, the expression of Netrin-1increased, Netrin-1receptor Neogenin expression reduced.With the receptors that reduce, Netrin-1promote the survival of cancer cells as ligands, andcontinuing added value, which eventually led to increased the level of the mechanism oftumor formation. In short, these results show that Netrin-1to express and Netrin-1and thereceptor Neogenin interaction, regulating tumor cell survival and increase of tumor grade, for further in the happen and development of human glioma, and provide some theoretical basisin the research of the mechanism and antitumor medicine development of glioma.
