| Objective:To explore the preventive protection effect of1,25-dihydroxyvitamin D3on myocardial apoptosis in rats with type2diabetes mellitus.Methods: Thirty-four maleSD rats were randomly divided into control group with8rats,type2diabetesmellitus(T2DM)group with12rats and intervention group(type2diabetes mellitus ratsintervened with1,25-dihydroxyvitamin D3) with14rats.After high-glucose and high-lipiddiet for6weeks,the intervention group was given1,25-dihydroxyvitamin D3lavage,andT2DM group was given peanut oil in the same dose as1,25-dihydroxyvitamin D3used inthe intervention group.Control group was fed with normal diet.The plasma glucose,bodyweight, cardiac weight,cardiac weight index, lactic dehydrogenase and creatine jubasewere detected,and morphology of cardiac tissues was observed after6weeks oftreatment,while the expressions of Fas and FasL in cardiac tissues were observed withimmunohistochemistry and RT-PCR in these three roups.Results: The levels of plasmaglucose, cardiac weight index, lactic dehydrogenase and creatine jubase were higher,andthe body weight,and cardiac weight were lower in T2DM and intervention group thanthose in control group(P<0.05).The levels of plasma glucose, body weight, lacticdehydrogenase and creatine jubase were lower in intervention group than those inT2DM(P<0.05). Cardiac weight and cardiac weight index had no significant differencebetween intervention group and T2DM group(P>0.05).The expressions of Fas and FasLwere higher in T2DM group than those in the other groups(P<0.05).Conclusion:1,25-dihydroxyvitamin D3can protect cardiac of diabetic rats by inhibiting the expressionlevels of Fas and FasL,and restraining myocardial apoptosis. |
