At present, the safety of the drug problem has become people always the focus of attention, ensurepharmaceutical quality is an important significance to ensure the broad masses of the people’s safe,effective and maintain people’s health.Therefore, research a fast and accurate, simple and non-destructivedetermination of the means of identification, become a new direction for the development of modernpharmaceutical analysis method. With the rapid development of laser technology, the weak signal detectiontechnology and computer application, the application of Raman spectroscopy obtained the breakthrough.Raman spectroscopy has many characteristics, for example the test sample non-contact, non-destructive,high sensitivity, short measurement time and small amount of samples, can be realized rapidnon-destructive analysis of samples; At the same time, sample will not caused by chemical, mechanical,and photochemical decomposition in the process of analysis, this analysis method is one of the hot researchfields of science analysis, has a unique advantage in pharmaceutical applications, has gradually developedinto a green analytical technique in the study of drug testing. As a powerful tool analysis to test substances,Raman spectroscopy combined with multivariate calibration of chemometrics and model identificationmethod, could obtain the maximum in the Raman spectra data and information, realize the online or offlinefor determining the content of tablets active component. So Raman spectroscopy and chemical metrologymethod can effectively improve the accuracy and stability of the analysis results to reduce the analysis time,with a strong practical.This topic combined with partial least squares of chemometrics for research drug component contenton the basis of Raman spectrum analysis technology, through the comparison between the measured valuewith Pharmacopeia method, finally to establish a rapid non-destructive quantitative analysis method fordetermination of drug content, and the method of reliability evaluation to prove the feasibility of thismethod provides a new method for the analysis of drugs testing, the Raman spectrum analysis method inthe field of pharmaceutical analysis has a more broad prospects for development. This paper researchcontent mainly includes two parts:The first part of single ingredient in the drug content determination was investigated. Technology to establish a quick determination of acetaminophen tablets of acetaminophen content of quantitativeanalysis method using Raman spectroscopy and partial least squares method. Original Raman spectrumafter the first-order derivative preprocessing, in405-971+1104-1676+2861-3118cm-1spectral range,choose the best number of principal components for10, using partial least squares method to establish thecontent determination of paracetamol in the samples of the optimal quantitative calibration model, themodel of the correlation coefficient is0.99947, RMSEP and RMSECV were0.144,3.88, shows that themodel has good robustness. The model predicted values and the measured values of the standard methodon t-test, P>0.05, visible quantitative information obtained by this method is ideal. Also the model for thereliability evaluation, good precision and recovery, proved the feasibility of this method. So Ramanspectroscopy combined with partial least squares method can be implemented for acetaminophen tabletsnon-destructive quantitative analysis.A quantitative study of the active ingredient in the second part of the multi-component drugs. Incompound paracetamol tablets of Raman spectra as the carrier, the partial least squares method is adoptedto process original effective information of spectra and extract, to establish a rapid determination ofcompound acetaminophen tablets of acetaminophen and aspirin the content of the quantitative analysismethod.. Original Raman spectra using the second derivative+smoothing spectral preprocessing methods,within the range of2550-3783cm-1, choose the best principal component number10, the partialleast-squares regression to establish the quantitative model, the model of the evaluation indicators havereached the best, shows that the model has good robustness and the ability to predict. Model forecastvalues compared with the measured value of the standard method, P>0.05, shows that the model compoundacetaminophen tablets of acetaminophen and aspirin content prediction results, and there is no significantdifference between measured value. Good precision and recovery rate, proves the feasibility of this method.Therefore, Raman spectroscopy combined with partial least squares method to realize the combination ofacetaminophen tablets non-destructive quantitative analysis, and the samples without pretreatment, fast andconvenient, has a great application prospect.
