ObjectiveTo investigate the expression and the significance of T-cadherin and soluble AdipoR indifferent parts of non-alcoholic fatty liver diseases (NAFLD) in SD rats.Methods1、 The establishment of model:A total of60SD rats were divided into2groups randomlyafter1weeks of normal feeding. Rats in a model group were fed with high-fat diet andrats in a control group were normally fed.2、 Check out index: Six rats of each group were killed at0,6,8,12,and16weeks. To observethe change in body weight and liver index, the level of fatty deposition in liver wasobserved using hepatic tissue pathological section(HE staining and SudanⅣ staining)and score of NAS(NAFLD activity score).T-cadherin mRNA expression and its dynamicchanges in the liver and blood were measured by RT-PCR. AdipoR1and AdipoR2mRNAexpression in the blood serum were measured by RT-PCR.Immunohistochemical assaywas used to detect the expression and dynamic changes in T-cadherin in the liver tissues.ELISA was used to detect the expression and dynamic changes of Adiponectin in theblood serum.Results1、 the change of liver index in each group: In the model group,the liver index was graduallyincreased, which was higher than that in the control group after6weeks(P<0.05).2、 Pathological changes of liver in each group: in the control group,there was no fattydegeneration in hepatic cells, and the hepatic tissue’s frame were integrity andclearness. In the model group,the liver were obviously enlarge and had fattydegenerat in different level after6weeks.The score of NAS was gradually increased, which was higher than that in the control group after6weeks(P<0.05).3、 the serum adiPonectin’s change: In the model group, the serum adiponectin was graduallydecreased, which was lower than that in the control group after8weeks(P<0.05).4、 the expression and changes of T-cadherin mRNA in hepatic tissues and blood:In the modelgrouP, the expression of T-cadherin mRNA in hepatic tissues and blood was graduallyincreased, which was higher than that in the control group after6weeks(P<0.01). Therewas significant positive correlation between T-cadherin mRNA expression in liver and liverindex (r=0.900,P<0.01)or score of NAS(r=0.933,P<0.01). Meanwhile, soluble componentsof T-cadherin were found in blood, which were positively correlated with the liver index(r=0.805,P<0.01) or score of (r=0.889,P<0.01).5、 the expression and changes of T-cadherin in hepatic tissues:In the model group, theexpression of T-cadherin protein in hepatic tissues was gradually increased, which washigher than that of the control group after6weeks(P<0.05). There was significant positivecorrelation between T-cadherin protein expression in the liver tissues and liver index(r=0.879,P<0.01)or the score of NAS(r=0.912,P<0.01).6、 the expression and changes of AdipoR1and AdipoR2:soluble components of AdipoR1were found in blood,which was the same with the two groups(P>0.05)。Meanwhile,AdipoR1was found in hepatic tissues, which was the same with the two groups(P>0.05)。In the model group, the expression of AdipoR2in hepatic tissues was gradually increased,which was higher than that of the control group after6weeks(P<0.05).Conclusion1、The expression of T-cadherin mRNA and protein in hepatic tissues and blood was positivecorrelatied with inflammation degree of liver in Nonalcoholic Simple Fatty Liver Diseaserats,suggesting that T-cadherin expression may be closely related to the pathogenesis ofNonalcoholic Simple Fatty Liver Disease.2、Solubility AdipoR1and T-cadherin were expressed in blood,and the expression ofAdipoR1and T-cadherin mRNA was positive correlatied with inflammation degree ofliver in Nonalcoholic Simple Fatty Liver Disease rats,suggesting that adiponectin and AdipoR may be closely related to the pathogenesis of Nonalcoholic Simple Fatty LiverDisease.3、In the model group, the expression of AdipoR2mRNA in hepatic tissues was graduallydecreased, suggesting that AdipoR2may be a negative regulator of Nonalcoholic SimpleFatty Liver Disease.
