Objective:To investigate the effects of liver function and liver regeneration when rats withobstructive jaundice hepatectomy using recombinant human growth hormone, and toprovide a theoretical basis for clinical application.Materials and Methods:1.Animal groups:72healthy male SD rats (purchased from Kaifu District,Changsha City, Hunan products biological limited liability company), weight320±20g, keeping a clean environment, SD rats were free to be fed on food and water andthe rhythm of life was12hours a day. The rats were randomly divided into fourgroups:1.SN(SO-NS) group: the rats with sham-operated after hepatectomy using theNS injection.2.SG(SO-rhGH) group: the rats with sham-operated after hepatectomyuseing the rhGH injection.3.OJ(OJ-NS) group: the rats with Obstructive Jaundiceafter hepatectomy using the NS injection.4.OG(OJ-rhGH) groups: the rats withobstructive jaundice after hepatectomy using the rhGH injection.2. Preparation of animal model:(1) SN group and SG group:10%chloral hydrate0.3ml/kg anesthetized by intraperitoneal injection, supine position, limbs fixedabdominal skin preparation, iodophor disinfection, abdomen was closed after followthe linea alba into the belly, find the common bile duct, be turning the first hepaticportal.5days later, freed left hepatic lobule,hypodesis left hepatic pedicle, thenhepatectomy,biliary tract drainage, then abdomen was closed. SN group to be dailysubcutaneous injection1ml/kg saline., SG group be the daily subcutaneous1ml/kgrecombinant human growth hormone.(2) ON group and OG group:10%chloralhydrate0.3ml/kg anesthetized by intraperitoneal injection, supine position, limbsfixed, abdominal skin preparation, iodophor disinfection, abdomen was closed after follow the linea alba into the belly, find the common bile duct, be0#silk ligature.5days later, free left hepatic lobule, hypodesis left hepatic pedicle, thenhepatectomy,biliary tract drainage, then abdomen was closed..SN group to be dailysubcutaneous injection1ml/kg saline, SG group be the daily subcutaneous1ml/kgrecombinant human growth hormone.3. Indicators detection: each phase of six SD rats in each group respectively,Postsurgical24h,72h,120h, postoperative residual liver weight and body weight ofrats was measured the rate of liver regeneration; automatic biochemistry analyzerdetect serum total bilirubin (TIBL), alanine aminotransferase (AST), albumin (ALB);remnant liver HE staining, use of light microscopy observated liver tissue mitosis,statistics on the number of mitotic cells, the mitotic index,immunohisto chemicalstaining of PCNA expression.Results:1. Postsurgical liver function testThe rat which with no obstructive jaundice underwent liver resection after24h,72h,120h, the SG group ALT declined faster than the SN group ALT, P <0.05, thedifference was statistically meaningful; postoperative24h,72h, SG group ALB morethan the SN group ALB (P <0.05), the difference was statistically meaningful. Ratpartial liver resection after biliary obstruction, in the24h,72h, SG group TBIL lessthan the SN group TBIL (P <0.05), the difference was statistically meaningful;24hafter surgery,72h,120h SG group ALT is lower than the SN group ALT, and the SGgroup ALB more than the SN group (P <0.05), the difference was statisticallymeaningful.2. Postsurgical liver regeneration evaluationSG group more than SN group postsurgical24h,72h,120h, the rate of liverregeneration, liver mitotic phase, P <0.05, the difference was statistically meaningful;SN group more than ON group postsurgical24h,72h,120h of liver regeneration rate,hepatocyte nuclear division (P <0.05), the difference was statistically meaningful; OGgroup more than ON group postsurgical24h,72h,120h, the rate of liver regeneration,liver mitotic (P <0.05), the difference was statistically meaningful; the OG group less than the SG group after24h,72h,120h, liverregeneration rate, liver mitotic (P <0.05),the difference was statistically meaningful.Conlusions1. Recombinant human growth hormone can significantly improve obstructivejaundice liver resection liver function in rat;2.Recombinant human growth hormone can promote obstructive jaundice afterhepatectomy residual liver regeneration, promote liver regeneration ahead of the peak.
