ObjectiveTo explore the effects of IL-7on the peripheral Th17/Treg cells and the pregnantoutcomes in murine RSA models in the period of implantation.Methods1)Female CBA/J mice were mated with male Balb/c and DBA/2micerespectively to construct normal pregnant and spontaneous abortion models.2) After overnight cohabitation with DBA/2or Balb/c males,CBA/J females withvaginal plugs were separated from the males and the day of vaginal plug detection wasconsidered to be day0of pregnancy.3) On the4th day of pregnancy (the period of implantation), DBA/2-matedfemales were then randomized and divided into two groups: one group injected15μl permice intraperitoneally, every other day, three times. The other group and the normalpregnancy group did not inject any drugs.4) On14th day of pregnancy, the females were sacrificed, We calculated the ratioof abortions.5) Pick up lymphocytes of CBA/J females in three groups.6) Analyzed the percentage of CD4+CD25+Foxp3+Treg and theCD4+IL-17A+T cell in spleen cells by flow cytometry instrument, to investigatepossible role of IL-7inducing Th17/Treg imbalance in murine RSA models. 7) By RT-PCR methods for the expression of Foxp3mRNA and ROR γt mRNAof samples. Explore whether IL-7adjust the Th17cells specific transcription factorRORγt and Treg cells specific transcription factor Foxp3and in murine RSA models.Rsults:1) The normal pregnant and spontaneous abortion models were successfullyconstructed.2) Abortion group treated with IL-7showed a significantly higher abortion ratecompared with the normal pregnancy group and the abortion group(P<0.05).3) The proportion of CD4+IL-17A+T cells in abortion group treated with IL-7(IL-7group) showed a significantly higher than the normal pregnancy group and theabortion group(P<0.05).However, The proportion of CD4+CD25+Foxp3+T cellS inabortion group treated with IL-7(IL-7group) showed a significantly lower than thenormal pregnancy group and the abortion group(P<0.05)4) RORγt mRNA expression was significantly increased in abortion group treatedwith IL-7(IL-7group) compared with the normal pregnancy group and the abortiongrou(pP<0.05). Foxp3mRNA expression was significantly decreased in abortion grouptreated with IL-7(IL-7group) compared with the normal pregnancy group and theabortion group(P<0.05).Coculsion:The normal pregnant and spontaneous abortion models were successfullyconstructed.Compared with the abortion group, the proportion of CD4+IL-17A+T cellsand the Th17cells specific transcription factor RORγt mRNA were both increased inabortion group treated with IL-7(IL-7group).These prompted that IL-7is essential forsurvival and expansion of pathogenic Th17cells. Treg may benefit the maintenance ofpregnancy. The expression of Th17cells was higher, Treg cells was lower in URSApatients.The balanced of Th17and Treg may be migration,it can increase theoccurrence of abortion.
