Objective Carcinogenesis and development of Lung cancer accompanied with thechanges of amino acids and elements levels. In this study, the serum/tissue free aminoacids and elements (As, Se, Cd, Cr, and Pb) in cancer of the patients with lung cancerwere determined, compared that with the health controls or paracancerous tissues, toanalyze the the possibility of amino acids and elements for clinical early-stage diagnosisand treatment of lung cancer.Methods (1) Amino acids:17pairs of Lung cancer tissues and paracancerous tissues,15serum samples of lung cancer patients were sampled from the Affiliated Hospitals ofMedical University,16healthy control serum samples were sampled from the PhysicalExamination Center of the same hospital. The free amino acids content were alldetermined by A300amino acid analyzer. The results were analyzed by WilcoxonSigned Ranks Test or Kruskal-Wallis H Test according to their statistical request.(2)Elements:7pairs of lung cancer tissues and paracancerous tissues were taken from thesame hospital. As and Se elements were determined by atomic fluorescencespectrometry; Cd, Cr and Pb were detected by inductively coupled plasma massspectrometer (ICP-MS). The results of the elements in lung cancer/paracanceroustissues were recorded by the mean and standard deviation. A meta-analysis wasconducted to analyse the relationship between lung cancer and five elements from the publications related to this field in the recent years.Results (1)22kinds of amino acids have determined in17pairs of lung cancer tissuesand paracancerous tissues, the content of Lys, Tau and NEAAs in lung cancer tissueswas significantly higher than those in paracancerous tissues (P<0.05). Glu content wassignificant difference between lung cancer tissues and paracancerous tissues(P<0.01).The content of NEAAs in cancer tissues was significantly increased by50.93%compared with paracancerous tissues.23kinds of free amino acids havedetermined in15groups of lung cancer patients’ serum and16groups of healthycontrols’ serum. Relatived to the healthy groups, Only the Phe and Hylys increasedsignificantly in lung cancer patients serum (P<0.05). Meanwhile, the content of Met,His, Val, Tau, Gly, Ala, Cit, α-ABA, Cys and Pro had a significant reduction in lungcancer patients serum (P<0.01). The content of EAAs was no difference between thetwo groups, but the content of NEAAs was significantly decreased by35.80%to thehealthy controls. BCAAs significantly decreased by20.49%, there was no change ofAAAs content, but the ratio of BCAA/AAA decreased significantly and decreased by29.20%to the healthy controls.(2) The content of Cr in lung cancer tissues andparacancerous was the highest, Se content was the lowest. The results of Meta-analysisshowed that the Cd content in the lung cancer tissues compared with paracanceroustissues reduced0.25μg/g,(95%CI:-0.47,-0.03; P=0.03), Se content in the lung cancertissues compared with paracancerous increased0.17μg/g,(95%CI:0.01,0.32; P=0.03).Conclusions The pattern of free amino acid between groups of lung cancer tissuesand paracancerous tissues were different. Glu content of the lung tissues significantlyhigher than the paracancerous, and the content of Lys、Tau and NEAAs was higer thanthose in the paracancerous tissues too. The pattern of free amino acid in serum ofpatients with lung cancer and the healthy controls was significantly difference too. Thecontent of Phe and Hylys was higer than the healthy group, the content of Met、His、Val、Tau、Gly、Ala、Cit、α-ABA、Cys and Pro were decreased to the healthy controls’; The variance of NEAAs、BCAAs、BCAA/AAA were significantly reduced. The contentof5elements in lung cancer tissues and paracancerous tissues were significantlydifferent: Cd content in the lung cancer tissues was lower than that in the paracanceroustissues, and the content of Se was higer than that in the paracancerous tissues.
