Objective:To research the effect of selective cycloxygenase-2blocker,celecoxib on XUAN WEI women adenocarcinoma of lung cell line(XWLC-05)and human adenocarcinoma of lung cell line(A549) in vitro, approach to find the possible mechanism and possible cure in XUAN WEI YUN NAN PROVINCE.Methods:selective cycloxygenase-2blocker, celecoxib in different range of concentrations were treated with these two kinds of lung adenocarcinoma cell lines(XWLC-05and A549) as single agents.Observe the change of cell appearance and growth state. Cell’s growth inhibition were measured by MTT. The cell cycle and apoptosis of these two kinds of cell lines were measured by flow cytometric analysis after celecoxib treated with the concentration of15μmol/L and30μmol/L. Observe the appearance of cell and apoptosis by fluorescence microscope. DNA Ladder testing Kit were used to observe these two kinds of cell lines’DNA Ladder after celecoxib treated.Results:(1) The growth of XWLC-05and A549can be inhibited by celecoxib, such effect showed a concentration-dependent and a time-dependent manners in a certain range. (2) Celecoxib significantly induced G0/G1(P<0.01) arrest and apoptosis (P <0.005) in XWLC-05and A549cell lines.(3) Treated with celecoxib, the apoptosis of XWLC-05and A549cell lines were observed in fluorescence microscope, DNA ladder were found in these two kinds of cell lines.Conlusion:In this study, we observed that blocking COX-2by celecoxib can inhibit the growth of XWLC-05and A549cell lines.With the inhibit of COX-2,that XWLC-05and A549cell lines were significantly inhibited in G0/G1arrest and produce cell apoptosis. COX-2inhibitors may become the new targeting therapy of non-small cell lung cancer in XUAN WEI YUN NAN PROVINCE.
