[Objective]1. Through the DVT rat venous tissue pre-genetic testing, screening differentially expressed genes and using Ncbi Gene bioinformatics platform, locking upstream genes COX-1which highly correlated with platelet aggregation.2. PCR detection COX-1gene expression in human blood.3. Biometric information and a Ncbi Gene Pathway analysis method to analyze the relation between COX-1gene and enhanced platelet aggregation and coagulation.It can be have more targeted research molecular mechanisms of the DVT occurs and development,and provide experimental basis for predicting the diagnosis of DVT molecular markers.[Material and method]1. Through build a rat model of DVT,detection differences in gene expression in femoral vein wall tissue by Genechip, combined Ncbi Gene, Pathway analysis, locking procoagulant gene COX-1gene as a research; Use biological information database platform, retrieved rats, humans COX-1Notes,through BLAST alignment to determine the COX-1gene between rats and humans Homology.2. It is collected161cases from March2011to March2013, strictly follow the DVT diagnostic criteria and inclusion and exclusion criteria (Appendix1), and ultimately enrolled in the study60cases divided into:normal control group (20cases), non-thrombosis group (20cases), thrombosis group (20cases).The three groups were collected fasting venous blood samples, non-thrombosis group in operation after1d acquisition, thrombosis group diagnosed after1d acquisition.Three groups of corresponding time points are simultaneously gater vein blood samples to detect routine blood,blood coagulation and D-dimer determination. COX-1gene expression was detected by PCR quantitative and Pathway bioinformatics analysis, analysis of the interaction between COX-1and coagulation system, platelet aggregation process of the formation of DVT.3. Using Pathway Analysis and bioinformatics methods to analyze between COX-1gene and the coagulation system, platelet aggregation on the formation of DVT.[Results]PCR detection of three groups of blood cells, the expression of COX-1results suggest that:three groups were expressed, Compared to the normal control group and non-thrombosis group, the thrombosis group was highly expressed COX-1have difference statistically significant (P<0.05), non-thrombosis group and normal control group showed no significant difference (P>0.05).[Conclusion]COX-1gene expression in three groups of human blood, more expression in patients with DVT, which may be involved in the formation of deep vein thrombosis in patients with DVT. |
