Effects Of Chinese Medicine "KuiLingJuaJiJian" On The Protein Expression Of Bax And Bcl-2in H22Hepatocarcinoma Cells Bearing Mice

Objective:To study the inhibitory effects of Chinese Medicine “KuiLingHuaJiJian”on tumor growth of transplanted hepatocarcinaoma in mice and expressionsof Bcl-2and Bax pooteins in tumor tissues.Method:Forty healthy Kunming mice,transplanted by H22hepatocarcinomacells,were divided into4groups,10mice per group,including Chinese medicinetreatment group, NS control group,5-fluorouracil(5-FU) treatment group andcombination of Chinese tradiational and Western medicine group. Chinese medicinetreatment group were given daily by mouth to Chinese medicine.Normal saline bymouth was given to the NS control group.5-FU by injection every day were given to the5-FU group.Combination group were given both Chinese medicine by mouth and5-FUby injection.15days later,all mice were sacrificed.The size of tumor tissues weremeasured and tumor inhibition rate was calculated.And the levels of theBcl-2and Baxprotein expressions were measured by immunohistochemistry.Result:The tumor inhibition rate of5-FU group and combination group were75.13%,52.06%,respectively,and were higher obviously than that in NS controlgroup(P<0.01).compared to the NS control group,expression of Bcl-2protein increasedand expression of Bax protein decreased in the groups of Chinese medicine treatmentand combination group(p<0.01).Conclusion:Apoptosis is initiative process of cell suicide that programmed bygenes.Apoptosis process is in connection with Bcl-2family,which play an importantrole on the regulation of apoptosis.According to the function, Bcl-2family can beclassified into two categories: one is represented by Bcl-2,which prevents apoptosis,and the other is represented by Bax and Bad,which promote apopto sis.Ku iLingHuaJiJiancould inhibit the growth of tumor,reduce the expression of Bcl-2protein and raise theexpression of Bax.KLHJJ has significant anti-tumor effects in vivo for inducingapoptosis of H22tumor cells.
