| Objective: Follistatin is an extracellar antagonist of BMPs’signaling. Follistatin isa combination of BMPs to BMPs receptors to hinder the BMPs to their own receptorsand downstream signal transduction through the formation of a trimer structure, whichplay a negative regulator of the role of BMPs into bone.The main purpose of this studyis to examine the expression of Follistatin in periodontal tissues during orthodontictooth movement in rats.Method: Selected218-week-old male SD rats(200±20g),which were randomlydivided into7groups.According to the classical cycle of orthodontic,we set up for the1day group,3days group,5days group,7days group,14days group,21days group,28days group.Establishing the orthodontic tooth movement model in SD rat oral.Theexperimental side of the left maxillary arch,the right to control side.A coil spring wasplaced between the maxillary first molar and the maxillary mini-implant,themini-implant are the anchorage teeth. The first molar was moved mesially by about40gforce.In accordance with the experimental group were the1,3,5,7,14,21,28days in theexperimental perfusion rats were killed and specimens were internal and enternalfixation.HE conventional histological staining,using streptomycin avidin-biotincomplex (SABC) immunohistochemical method to detect ratsduring orthodontic toothmovement in Follistatin in periodontal tissue expression.PBS instead of primaryantibody as negative control.Analysis software with spps13.0gray value of themeasured and analyzed statistically.Results: Follistatin has a specific temporal and spatial expression profiles duringthe orthodontic tooth movement. The expression of Follistatin was clearly upregulatedin the early phase and then decreased thereafter. It reached the peak on7day,then decreased gradually,the expression is almost equal to the control group on28daygroup.Conclusion:In the tension side of the periodontal membrane and alveolar boneosteoblasts and fibroblasts positive expression of Follistatin shows that Follistatininvolved in the rat bone remodeling and orthodontic tooth movement. As anantagonist,follistatin often is expressed in conjunction with BMPs,conceivably definingboundaries of BMP induced structures.The upregulation of the Follistatin expressionpoints to the possibility of a negative feedback mechanism for BMPs induced boneformation. |
