Objective: To study the expression level of the NLRP3inflammasome mRNAand Caspase-1mRNA in periphery blood monocytes (PBMCs) and the associated factorin periphery blood plasma from patients with coronary atherosclerotic heart disease andto explore the effect of NLRP3inflammasome in coronary atherosclerotic heart disease.Methods: Blood samples were respectively collected from18cases of healthypeople,15cases of the stable angina pectoris (SA) and38cases of acute coronarysyndrome (ACS),including18cases of the unstable angina (UA) and20cases of acutemyocardial infarction (AMI). We collected6mL blood from radial artery to get themonocytes and the plasma, Among these AMI patients’ blood was collected in thephase of6hours from its onset. Using real time-PCR (RT-PCR) to investigate theexpression level of the NLRP3mRNA and Caspase-1mRNA in PBMCs which frompatients with healthy people, SA and ACS. Using enzyme linked immunosorbent assay(ELISA) to explore the IL-1β and IL-18in plasma. Then use one-wayANOVA tocompare the difference of these indicator in each group, and analyze their correlationwith NLRP3mRNA. Use Spearman’s correlation to evaluate the relationship betweenNLRP3mRNA and Gensini score, GRACE score, CK-MB, cTnI, and LDL.Results: The expression of NLRP3mRNA in SA(1.035±0.167) and in ACS(1.676±0.189) were significantly higher than the controls(0.756±0.101)(p<0.05), whilethe expression of NLRP3mRNA in ACS increased significantly when compared to SA.The expression of Caspase-1mRNA in SA(2.027±0.058) and in ACS(2.009±0.116)were significantly higher than the controls(1.781±0.051)(p<0.05), but there was nosignificant difference between SA and ACS(p=0.824,p>0.05). The concentration ofIL-1β in SA(1.033±0.195) and in ACS(2.269±1.823) were signifycantly higher than thecontrols(0.461±0.119)(p<0.05), while the concentration of IL-1β in ACS increased significantly when compared toSA. The concentration of IL-18in SA(115.561±7.543)and in ACS(208.303±13.225) were significantly higher than the controls(67.922±18.405)(p<0.05), while the concentration of IL-18in ACS increased significantly whencompared to SA. Using Spearman’s correlation,we found signifycant correlationbetween NLRP3mRNA and Caspase-1mRNA (r=0.517,p=0.00), the IL-1β(r=0.313,p=0.008),and IL-18(r=0.413,p=0.00). In ACS patients, Spearman’s correlation analysisshowed no significant correlation between Gensiniscore(r=0.16,p=0.092), GRACEscore(r=0.14,p=0.063),CK-MB(r=0.06,p=0.738),cTnI(r=0.13,p=0.438),andLDL(r=0.11,p=0.346).Conclusion: Dysregulated of expression of the NLRP3inflammasome inperiphery blood plasma from patients with coronary atherosclerotic heart disease mayplay an important role in the process of coronary artery inflammation.
