objective:This study aimed to observe the differences in the levels of immune cell,specially with lymph cell subjects and CD4+CD25+CD127lowregulatory T cells(Treg)and the other related immunocyte subsets in the peripheral blood, and TSGF and sIL-2R in plasma between the healthy controls and the patients with gastric cancer. To investigate the relationship of these differences with the clinical pathological conditions and the responding clinical significances.Methods:The levels of related immunocyte subsets include Treg cell and sIL-2R and TSGF in the peripheral blood in50healthy subjects and137patients with gastric cancer,were detected by FCM and ELISA. In addition, the informations of the clinical and pathological conditions were collected, and the correlation were anyalzed with the above markers.Results:1. The patients with gastric cancer had a higher levels of CD3+T cell compared with the healthy controls.[70.4891±10.36%(n=137)and69.7820±6.90%(n=50), P>0.05]The patients with gastric cancer had a lower levels of CD4+T cell compared with the healthy subjects.[37.1876±7.98%(n=137) and39.3540±6.38%(n=50), P>0.05]2.The levels of CD8+T cells in the gastric cancer patients’peripheral blood, were significantly higher than those in the healthy subjects [30.5489±11.35(n=137)and24.8180±6.18%(n=50)(P<0.05)].3. The ratio of CD4+/CD8+in the gastric cancer patients’peripheral blood were significantly lower than those in the healthy subjects [1.2015±1.92%(n=137)and2.2064±3.54%(n=50)(P<0.05)]4. The levels of Treg cells in the gastric cancer patients’peripheral blood were significantly higher than those in the healthy controls.[7.0496±2.41%(n=137)and6.1120±1.22%(n=50)(P<0.05)].5.The levels of sIL-2R in the gastric cancer patients’peripheral blood, were significantly higher than those in the healthy subjects [345.3577±100.07u/ml (n=137)and326.4523±95.75u/ml (n=50)(P<0.05)]6.The levels of TSGF in the gastric cancer patients’peripheral blood, were significantly higher than those in the healthy controls [66.3285±9.29u/ml (n=137)and58.3175±6.21u/ml (n=50)(P<0.05)].7.The levels of NK cells in the gastric cancer patients’peripheral blood were significantly lower than those in the healthy subjects [5.8577±4.22%(n=137)and9.9600±4.26%(n=50)(P<0.05)]. 8. The levels of NK cell in the gastric cancer patients’peripheral blood were decreased with TNM classification, stage0+I (n=17) NK cell level was7.8357±4.42%, stage Ⅱ (n=23) was5.4000±3.78%, stage Ⅲ (n=82) was5.5575±4.22%, stage Ⅳ(n=15) was4.6125±2.76%, healthy control was9.9600±4.26%(n=50), stage Ⅱ、Ⅲ、Ⅳ IVwere significantly lower than those in the healthy subjects.9. The levels of Treg in the peripheral blood was higher in the patients with metastasis of lymph node than that in the patients without metastasis of lymph node, but not significantly [7.5425±2.53%(n=99)and6.8427±2.35%(n=38), respectively,P>0.05].10. The levels of TSGF in plasma in the patients was significantly higher than that in the patients without metastasis of lymph node[67.5455±9.24(n=99)and63.1579±8.75(n=38)(P<0.05)]。11. The levels of sIL-2R in plasma in the patients was significantly higher than that in the patients without metastasis of lymph node[351.2626±87.80u/ml(n=99)and329.9737±126.69u/ml(n=38)(P<0.05)]。12. The levels of Treg in the patients’peripheral blood were positively correlated with the TNM classification in the patients with gastric cancer(P<0.05). As the TNM classification advanced, the level of the Treg in the patients’peripheral blood was increased. The levels of plasma TSGF and sIL-2R in the gastric cancer patients were positively correlated with the TNM classification (r=0.219, P=0.011and r=0.19, P=0.028respectively, P<0.05). As the TNM classification advanced, the level of the plasma sIL-2R and plasma TSGF in the peripheral blood was increased.13. The levels of NK cells in the peripheral blood were negtively correlated with that of Treg cells in the patients with gastric cancer.(r=-0.203,P<0.05), The levels of NK cells decreased as Treg cells increased. The levels of sIL-2R in the peripheral blood has no correlattion with that of Treg in the patients with gastric cancer, The levels of Treg in the peripheral blood has no correlattion with that of TSGF in the patients with gastric cancer.Conclusion:1.The levels of CD3+T、CD4+T、NK cell and CD4+T/CD8+T in the peripheral blood of the patients with gastric cancer were lower than that of healthy control.And CD8+T cell were significantly higher than those of the healthy subjects. It can reflect the cellular immune function in those patients with tumor.But because CD4+T/CD8+T cell subjects contain Th/Ts cells, the results may not reflect the immune fuction in above.So dectect combine with Treg cell maybe well.2. The levels of Treg in the peripheral blood of the patients with gastric cancer were significantly higher than those of the healthy controls, and were correlated with the classifaction TNM pathological conditions. The levels of Treg in the peripheral blood were significantly correlated with the TNM classification in the patients with gastric cancer(P<0.05). Maybe tumor cell secret tunmor inhibit cell factor promote Treg cell differentiation and reproduce,3. The levels of Treg in the peripheral blood were negatively correlated with NK cells in the patients with gastric cancer,which implied that the Treg may inhibit the immune function by directly or indirectly regulation of the secretion of CD4+T cells and NK cells in patients with gastric cancer.4. The levels of sIL-2R and TSGF in the gastric cancer patients’ peripheral blood were significantly higher than those in the healthy subjects. The levels of plasma sIL-2R and TSGF in the gastric cancer patients were positively correlated with the TNM classification. As the TNM classification advanced, the level of the plasma sIL-2R and plasma TSGF in the peripheral blood was increased. It is suggested that the development of gastric cancer may relate with the suppression of immune function.5. The gastric cancer patients had significantly higher levels of Treg and CD8+T cells and sIL-2R、TSGF in the peripheral blood, and a significantly lower level of CD4+T cells in the peripheral blood than the healthy controls. If the levels of these immune cells and cytokines were combined detected in the patients with gastric cancer, that may reflect the cellular immune function more accurately. It is not only juge the the maglinant lever of the tumor, also can find the development of the subject from immune function.It wound be important in disease control and formulate of treatment.
