[Objective] This study aimed to investigate the protective effects of atorvastatinagainst opromide induced renal tubular cell apoptosis in diabetic rats.[Methods] Streptotocin-induced diabetic rats were fed for eight weeks and thenrandomly assigned into four groups as normal control group (N group),diabetic group (DM group), group of diabetic rats injected withdiatrizoate (DM+CM group) and diabetic rats respectively treated with5mg/kg/d atorvastatin (ATO1group),10mg/kg/d atorvastatin (ATO2group),30mg/kg/d atorvastatin (ATO3group) before iopromide injection.Urine creatinine (Ucr) and24hours urine albumin (24h-UAlb) weredetermined24hours after iopromide injection.Serum creatinine (Scr)and Blood urea nitrogen (BUN) were detected48hours after iopromideinjection, then creatinine clearance rate (Ccr) and urinary albuminexcretory rate (UAE) were computed.The rats were sacrificed and bothkidneys were removed48hours after iopromide injection, then for theleft kidney, morphology, renal tubular apoptosis byTdT-mediated-dUTP-nick-end labeling (TUNEL) and expression of Baxand Bcl-2in renal by immunohistochemistry were detected. For the rightkidney, expression of Bax and Bcl-2were measured with western-blot.[Results] Compared with N and DM group, the level of Scr, BUN and UAE,aswell as the expression of Bax in renal cortex were higher,the level ofCcr and Bcl-2activity in renal cortex were less and TUNEL-positivecells were more in DM+CM group. Compared with DM+CM group,atorvastatin groups ameliorated these changes, especially in30mg/kg/datorvastatin group. [Conclusion] Opromide could cause renal tubular apoptosis. Early application ofatorvastatin could dose-dependently ameliorate the development ofCI-AKI, might partly due to suppression of opromide induced renaltubular apoptosis.
