| Plant Pterocarya stenoptera C. DC.(Juglandaceae), alternately named as PingLiu, Ma Liu, Shui Ma Liu, Wu Gong Liu, is widely distributed in north and centralChina. It has been used as a traditional Chinese medicine for the treatment of variousdiseases, such as dispelling wind and eliminating dampness, decreasing swellingrelieving pain, antipyretic, and insecticidal activity. In the folk, Pterocarya stenopteraC. DC. is mainly used to treat various types of fungal-infected skin diseases andbacterial abscess. So far, the report on the chemical composition of Pterocaryastenoptera C. DC. is very few. In order to find the active compound to a deeperunderstanding of the chemical composition of the plant, we carried out a systematicstudy on the chemical constituents in the bark of Pterocarya stenoptera C. DC.. Thechemical constituents in chloroform and ethyl acetate fractions of90%ethanol extractfrom the barks of P. stenoptera C. DC. were isolated and purified by columnchromatography of silica gel, ODS, and Sephadex LH-20, preparative HPLC andrecrystallization. The structures of compounds were identified on the basis ofphysicochemical properties and spectral data analyses. Twenty compounds wereobtained from barks of Pterocarya stenoptera C. DC..12compounds were isolated andidentified from the chloroform fraction, and they are glochidone (1), lup-20(29)-en-1β,3β-diol (2), betulinic acid (3),3-O-acetyl-betulinic acid (4), ursolic acid(5), oleanolic acid (6),2α,3β,19α-trihydroxyurs-12-en-28-O-β-D-glucopyranoside(7),30-norlupan-28-oic acid (8), β-sitosterol (9),(4S)-4,8-dihydroxy-1-tetralone (10),5-hydroxy-4-methoxy-α-tetralone (11),(4S)-4-hydroxy-1-tetralone (12); Other8compounds were isolated and identified from the ethyl acetate fraction, and they aremyricitrin (13), quercetin-3-O-2″-galloyl-rhamnoside (14),myricetin-3-O-2″-galloyl-rhamnoside (15), myricetin (16),3,3′-di-O-methyl ellagicacid-4′-O-β-D-xylopyranoside (17),3,3′-di-O-methyl ellagic acid-4′-O-β-D-rhamnoside (18), quercetin (19), β-daucosterol (20). The compounds obtained fromthe barks of Pterocarya stenoptera C. DC. are main triterpenes, flavonoids and naphthoquinone, compounds1-8, and10-19are isolated from the plant for the firsttime, and compounds7-8, and10-18are obtained from genera Pterocarya for the firsttime. Compounds7and10exhibited potent inhibitory effect on Hep-2with50%inhibition concentration (IC50) value of6.7μg/mL and9.4μg/mL, respectively.. |
