| Obiective:To investigate the prevention of Paclitaxel and At-RA(All-trans retinoicacid)respectively and their associated effect on allograft angiopathy in rats, anddiscuss their potential mechaniam of action and possible synergistic action.Methods: The establishment of allograft model was used with carotid transplantationin rats.60rats were randomly classified into four groups:the control group(A),At-RA–treated group (B),Paclitaxel-treated group (C) and combination-treatedgroup(D).At0,1,4,8weeks after transplantation, the lipoprotein profile and NOcontentã€ALTã€Crin blood serum were detected, also the grafts were harvested toobserved the regional changes in the intimal layers by light microscopy and theexpression of TNF-αã€PCNAã€CD4+T cells in allografts by immunohistochemistry.Results: There was the least intimal thickness, the highest level of NO in blood serum,the least expression of PCNAã€TNF-α and the expression of CD4+T cells in groupD.The opposite is group A. And group B and C are better than groupA.(P<O.05)Conclusion: The prevention of Paclitaxel and At-RA on allograft angiopathy wassignificant at the dose of this experiment,and the effect was most obvious in acombination of both drugs.At-RA may exert its function to prevent allograftangiopathy by the mechanism of secretion of NO,indirect anti-inflammatory effect,commissural inhibition on the expresstion of TNF-αã€PCNA and CD4+T cells and theproliferation of SMC.... |
