Desoxyribonucleic acid (DNA) is the carriers of biologic geneticinformation. It plays a very vital role in upgrowth of creatures and operation ofphysiological function. The research of interaction between small molecular andDNA is of grate significance in illustrating the action mechanism of anticancerand antiviral, early pharmaceutical screening in vitro, the origin of disease andthe development of probes of DNA structure. And, it is important researchsubject in many fields, such as Life science, Chemical biology, Clinical medicineand so on.In this thesis we synthesized six Hematoxylin (HE) complexes, whose centre ionis Ag, Dy, Eu, Mg, Ni, Zn respectively. These complexes were characterized byinfrared spectrum, ultraviolet spectrum, fluorescence spectrum and ultimateanalysis. The interaction between HE-metal complex and Herring sperm DNA(hsDNA) was studied by ultra-violet and visible spectrum, fluorescence spectrumand viscosimetry method. The binding ratios of six complexes were determinedby molar ratio method in physiological pH environment (pH=7.40), and thecorresponding thermodynamic parameters and binding constants were calculated,which uncovered the prime driving force of the interaction process. Researchresults shows that all six HE-metal complex could partial intercalate into thebase pair of hsDNA, it causes kink and bend of double helix of hsDNA, and theviscosity decrease. Specially, HE-Ag+complex mainly intercalate into A: T basepair, there also exist electrostatic binding between HE-Ag+complex and hsDNA.For else five complexes, they can bind with hsDNA through the groove ofhsDNA.
