| Cultivars comparison test and production test for six F1hybrid combinationsbred in Hebei Normal University were conducted in solar greenhouse with twocommercially usedcultivars, LÇœfengwang and Guifeicui, as control. CombinationsD7and D4were selected according to the breeding objectives. With the strongbranching combination D7, then experiments of planting density and labor-savingcultivation technology were carried out. RAPD DNA fingerprinting were developed,which provided a basis for solar greenhouse cucumber production using newcultivars. The main results are as follows:1. Based on the results of solar greenhouse cultivar comparison and productiontest, D7was a preliminary advance selection with a high fruit quality, high yield andstrong branching, D7was thus suitable for labor-saving cultivation in the solargreenhouse. For D7, the plant growth vigor was medium and the plant type wascompact. Most fruit set on the main vine with a strong branching ability. The leaveswas in a smaller size, The fruit was about13cm long, the average weight was about154g. Fruit shape was short cylindrical, The fruit skin color was uniformly brightgreen, The pedicle section of fruit was short in dark green color. The flesh was thickin light green, tasted sweet and crisp with light fragrance. The vitamin C content ofD7was4.38mg/100g which was significantly higher than the two control cultivarsLÇœfengwang and Guifeicui. The fruit sugar/acid ratio and total yield of D7did notsignificantly differ from Guifeicui but was significantly higher than LÇœfengwang.2. In winter-spring crop season under the regular production condition with80/50cm large/small raw, the yield of advance selection D7was the higher whenplant spacing was25~30cm than35cm indicating the best planting spacing of D7was25~30cm.3. In autumn-winter crop season labor-saving cultivation experiments in solargreenhouse, the total yield of D7own-root plants was higher by18.33%and the rateof commercial fruit harvested was higher by5.05%in decapitation than vine-fall treatment. The labor cost was reduced by640yuan/666.7m2in decapitationcompared to vine-fall treatment, the total yield of D7plants grafted on rootstockswas higher by19.89%and the rate of commercial fruit harvested was higher by19.89%in decapitation than vine-fall treatment. The labor cost was reduced by800yuan/666.7m2in decapitation compared to vine-fall treatment, These provides atheoretical basis for autumn-winter season labor-saving cultivation in solargreenhouse.4. The DNA fingerprinting of D7and the two control cultivars LÇœfengwang andGuifeicui were established using RAPD molecular markers. Four markers, A104ã€H05ã€S331and S500, were screened from120RAPD primers. The4markersgenerated five polymorphic loci. Four similar cucumber cultivars can be identifiedby the fingerprinting results with high resolution(1/42000). |
