| Potted flower of Dendrobium nobile have high ornamental value and its flowering is consistent with Chinese traditional Spring Festival,so Dendrobium nobile is gradually becoming the new bestowed favor of flowers saled during the Spring Festival.Seedlings of Dendrobium nobile newly introduced from Japan by Zhejiang Senhe Seed Industry Co., Ltd. is insufficient and is in short supply.Tissue culture propagation with fast speed and has its unique advantages to resolve the insufficient seedlings in production. The research set up the rapid propagation system of Dendrobium nobile through clustered shoots and protocorm two ways and optimized formula in different stages in order to provide scientific basis for intensive production.Five good cultivars’Huoniao’ã€â€™T-0’ã€â€™T-3’ã€â€™818’ã€â€™920’ were selected to study the rapid propagation system of Dendrobium nobile. Main results are as follows:The better medium of inducing mixed buds of clustered shoots way is MS+TDZ0.10mg·L-1+NAA0.20mg·L-1;the optimal multiplication medium is1/2MS+TDZO.10mg·L-1+NAA0.20mg·L-1+mashed potatoes100g·L-1;The better culture conditions is pH5.4and red and blue light ratio of7:3;Adding AC lg·L-1can effectively overcome browning phenomenon appeared in the process of proliferation.The better induction of material for protocorm is1-2mm bud tips and the better induction medium is MS+TDZ0.05mg·L-1+NAA0.50mg·L-1; The better multiplication medium for protocorm is:1/2MS+TDZ0.05mg·L-1+NAA0.10mg·L-1+LH2g·L-1+ABA0.50mg·L-1and pH is5.8-6.2;The optimal plantlet differentiation medium is1/2MS+TDZ0.10mg·L-1+NAA0.20mg·L-1The optimal medium for plantlet rooting is1/2MS+IBA0.50mg·L-1+suger20g·L-1+mashed potatoes100g·L-1;The better time of opening sealing membrane and smelting plants is5days;Water moor and cornerstone volume ratio1:1is more appropriate to be as transplanting medium.The study of reducing the cost were carried out on proliferation phase of clustered shoots way and the results showed that tap water can be used to instead of distilled water as water source andï¿¥0.60can be saved per liter of medium on water costs;30g·L-1of white granulated sugar can be used to instead of30g·L-1sucrose and48.0%carbon costs can be saved;5g·L-1of AGAR is conducive to proliferation of ’Huo niao’ã€â€™T-3’ã€â€™818’ã€â€™920’ and16.7%coagulant costs can be saved.And4.5g·L-1of AGAR is conducive to proliferation of ’T-0’ and25.0%coagulant costs can be saved; The study showd that the appropriate area of proliferation of each Dendrobium nobile’Huo niao’ã€â€™T-0’ã€â€™T-3’ã€â€™920’ plantlet is6.64cm2and production costs of per plantlet dropped to60.0%. And the appropriate area of proliferation of each Dendrobium nobile ’818’ plantlet is8.30cm2and production costs of per plantlet dropped to75.0%. |
