Originated in tropical and subtropical regions, Volvaria volvacea played an important role in the production of edible fungi for its ability of fruiting in high temperature season and short growth cycle. However, people have little knowledge in the genetic background of V. volvacea, which limit the breed improvement progress in this species. V. volvacea has a narrow genetic base, which limited the development of polymorphic molecular markers and impeded the construction of genetic map as well as its applications. Here, we developed SSR markers basing on the whole genome sequence data, and constructed a genetic map for this species. The major results are listed as follows:Two cultivation strains, Ping You NO.1and V23, were used as mapping parents in this study. And molecular markers based on the sequence of mating type were used for homokaryon detection. Hybrized the homokaryons which had different mating type, and selected the hybrids by the mating type molecular markers and fruiting test. After the detection of mating type by molecular markers,96homokaryons derived from the hybrid PV19were collected and constructed the mapping population.1346SSR markers were developed basing on the V23-1genome sequence. Among these SSR,57.4%were mononucleotide,29.7%were trinucleotide,8.2%were dinucleotide, the other motif type occupied a small proportion. Analysed the4020primers’polymorphism in mapping parents with e-PCR,1367primers’ product had length variance, and1242primers’product length variance were small than30bp. Verified the result with658primers by PCR, most of the primers accorded with the analysis, which indicated the using of e-PCR could improve selection efficiency of polymorphic SSR markers.Screened the polymorphic SSR markers in mapping population, and constructed the genetic linkage map using JoinMap4.0. Among the123SSR markers,118SSR markers distributed in19linkage groups,5SSR markers were not linked with each other. The total length of the genetic linkage map was1287.9cM, and the average distance between two adjacent markers was10.8cM.This SSR genetic linkage map will play an important role in gene mapping, QTL analysis, molecular breeding, physical mapping and comparative genomics research in V. volvacea.
