31varieties of Chrysanthemum morifolium has been taken for preliminary evaluation of genetic stability and quality which studied on morphological character and quality through a field experiment.1. The six multistate and fourteen quantitative characters of Chrysanthemum by coefficient variation and t-test by field experiments in the past2years. Results showed: most multistate characters were very coincident, and the quantitative traitses were affected greatly by environment factors.2. Suppling a supplementary means for find and identification the resistance cultivars of Chrysanthemum morifolium’Hangbaiju’leaf spot. The extent of genetic variation of21Chrysanthemum morifolium’Hangbaiju’materials was measured at the molecular level by random amplified polymorphic DNA (ISSR). The results show that21Chrysanthemum materials are clustered into3ISSR groups which is correlated with the resistance of Chrysanthemum to leaf spot. Showed that the ISSR technique can be used as an aid in screening the disease resistance cultivars.3. To provide basis for exploring the disease resitance mechanism. Diseased plants and diseased condition of the six varieties were recorded to calculate the average incidence and average index of disease condition. The disease resistance, the lignin content and activities of PAL and POD in series from strong to weak were NJK-2>NJK-1>NJK-3>CCh. morifolium’Hongxin11’> Ch. morifolium’Dahua4’> Ch. morifolium’Youdan6’, contents of total flavone were signidicantly different in different parts, contents of total flavone were all significantly different among the six varieties. Among the six varieties, the NJK-2were the strongest while Ch. morifolium’Dahua7’were the weakest; Contents of total flavone were all fairly higher in root, stem and leaf for the six varieties, while galiteolin mainly distributed in inflorescence.4. Analysis of chlorogenic acid, luteolin glycosides, iso-chlorogenic acid A, total flavone,volatile oil content and Chrysanthemum polysaccharide among31varieties of Chrysanthemum morifolium to make Comprehensive evaluation of Chrysanthemum. The result showed all test chemical component is significant different in31varieties, all variety types showed higher content of luteolin glycosides than Chinese Pharmacopoeia ediction2010, C. morifolium’Xinbaiju’×C. nankingense and C. morifolium’Hongxin8’showed lower content of chlorogenic acid than Chinese Pharmacopoeia ediction2010; C. morifolium’Xinbaiju’×D). indicum showed lower content of iso-chlorogenic acid A than Chinese Pharmacopoeia ediction2010.5. The content chlorogenic acid, luteolin glycosides, iso-chlorogenic acid A in different degrees of flower openness which of5varieties of Chrysanthemum were determined by using HPLC; essential oil was extracted by steam distillation; the content of Chrysanthemum polysaccharide was determined by spectrophotometric method. The study found that all variety types except C. morifolium’Sigong2’, the content chlorogenic acid, luteolin glycosides, iso-chlorogenic acid A and total flavone a maximum value appeared in70%degrees of flower openness, when was generally harvest period of Chrysanthemum, and the maximum value appeared in the time of Chrysanthemum Buds in C. morifolium ’Sigong2’, the test chemical component in5varieties of Chrysanthemum appeared the lowest value in100%degrees of flower openness. Chrysanthemu polysaccharide on different degrees of flower openness showed "7" shaped curve and reached the peaks during Chrysanthemum Buds period, the variation law of component essential oil had was the same. That showed the un-optimal harvesting time caneffect the content of chemical.6. To provide basis for exploring comprehensive development and utilization, the main chemical components distribution law of six Chrysanthemum morifolium’Hangbaiju’ varieties were comparatively studies. Contents of chlorogenic acid,3,5-O-dicoffeoylqunic acid and total flavone were signidicantly different in different parts, galuteolin content in inflorescence was the highest while no obvious difference of galuteolin content were found in root, stem and leaf; Contents of chlorogenic acid,3,5-O-dicoffeoylqunic acid and total flavone were all significantly different among the six varieties,but not for the galuteolin content. Conclusion:Contents of chlorogenic acid,3,5-O-dicoffeoylqunic acid and total flavone were all fairly higher in root, stem and leaf for the six varieties, while galiteolin mainly distributed in inflorescence.
