The collection and identification of cultivar resources in Chrysanthemum morifolium cultivated in BoZhou, AnHui Province was carried out:the plant in vitro propagation, plant growth in the field of Chrysanthemum morifolium cv.BoJu were studied, and the processing quality of BoJu flow was analyzed in contrast with the medicinal flos chrysanthemiof the other three clutivars.The main results achieved were as follows.1. Collection and identification of cultivar resourcesCollected resources colonization in Bozhou Chinese medicine research and development center Demonstration Park in Qiaodong town, Qiaocheng district, Bozhou, Anhui province. After the whole growth period, observation on plant morphological traits. Consistent with the characters of Chrysanthemum morifolium cv.BoJu in flora of China described by authenticated. It was the really Chrysanthemum morifolium cv.BoJu of Anhui medicinal plants.2. Chrysanthemum morifolium cv.BoJu in vitro propagationChrysanthemum morifolium cv.BoJu tip was taken as explants, the suitable medium of primary cultured was MS+6-BA0.5mg/L+NAA0.1mg/L, the formula of differentiation rate was92%.The bud seeding was strong and no callus.Stems shear was used to multiplication of bud clumps and transfer20days. The results showed that, with the increase of6-BA concentration, plants grow slowly, plant color gradually deepened, no significant difference in growth. MS medium was the suitable medium.Chrysanthemum morifolium cv.BoJu root easily. The results showed that,1/2MS+IBA0.3mg/L was the best medium of rooting of tissue culture seedlings of Chrysanthemum morifolium cv.BoJu. Rooted after5days and rooted11in average. Plant rooted developed and grown strongly.Transplanting after3-5days of Lianmiao period. The results showed that, soil:peat: perlite (1:1:1) was the best medium of transplanting of tissue culture seeding of Chrysanthemum morifolium cv.BoJu. The survival rate was100%and grown strongly.3. The growth condition in the field of Chrysanthemum morifolium cv. BoJuThe field observation, record and statistical analysis showed that:the survival rate of the original seedlings and the stock seedlings was100%. The comparative results between the stock seedlings and the CK group revealed that the yield of the stock seedlings improved by55.83%and had significant differences. The comparative results between the original seedlings and the CK group revealed that the yield of the original seedlings improved by28.05%and no significant differences. Compared with the original seedlings, the stock seedlings and the CK group, the weight of one hundred flowers increased of43.83%,30.53%respectively, the dry discount rate reduced by12.79%,12.37%respectively. The original seedlings and the stock seedlings of Chrysanthemum morifolium cv.BoJu had dark green, large leaves and crack depth, the plant diseases and insect pests to a lesser degree. The CK group had light green and small leaves, and there were many abnormal leaves such as floral leaves, yellow leaves. Compared with the original seedlings, the stock seedlings and the CK group, the wilt index decreased by48.2%,42.9%respectively, the septoria dipsaci index decreased by53.8%,44.%respectively, virus disease index decreased by66.7%,33.3%respectively, the aphid index decreased by42.1%,35.7%, and chrysanthemum gall midge index decreased by57.1%,42.9%.4. Study on processing methods of Chrysanthemum morifolium cv.BoJuStudy on processing methods of Chrysanthemum morifolium cv.BoJu. The results showed that, the content of chlorogenic acid, luteolin-7-O9-glucoside,3,5-O-discaffeoylquinc acid in Chrysanthemum morifolium cv.BoJu from drying the shade was highest, were0.58%,0.08%,2.64%respectively. The contents of luteolin-7-O-glucoside from different drying temperature cannot reach the minimum standards of Pharmacopoeia rules. The content of three effective component from microwave heating dryer and steam drying also cannot meet the minimum standards of Pharmacopoeia rules.5.Contrast test of tissue culture seedlings and regular seedlings of Chrysanthemum morifolium cv.BoJuAccording to the measured indicators of effective elements of Chrysanthemum morifoliumm pharmacopoeia2010edition of the people’s republic of china(Part one).Used HPLC to measure the content of chlorogenic acid, luteolin-7-O-glucoside,3,5-O-discaffeoylquinc acid in the original seedlings, the stock seedlings and the CK group. The results showed that, the content of three effective elements between the original seedlings and the stock seedlings similarity. The comparative results between the original seedlings and the CK group revealed that three effective elements improved by54.84%,100%,34.46%respectively. The comparative results between the stock seedlings and the CK group revealed that three effective elements improved by64.51%,200%,51.41%respectively. 6. Contrast test of quality of four kinds of medicinal flos chrysanthemiUsed HPLC to measure the content of three effective elements of BoJu flow and analyzed in contrast with the medicinal flos chrysanthemi of the other three clutivars. The results showed that, the content of chlorogenic acid and3,5-O-discaffeoylquinc acid was the highest, and had less luteolin-7-O-glucoside. The comparative results between Chrysanthemum morifolium cv.BoJu and GongJu revealed that chlorogenic acid and3,5-O-discaffeoylquinc acid improved by24.44%,138.80%respectively. And compared with HangbaiJu, the two effective elements can improve by47.37%,60.12%respectively. And compared with ChuJu, the two effective elements can improve by24.44%,174.26%respectively. The comparative results between Chrysanthemum morifolium cv.BoJu and GongJu revealed that luteolin-7-O-glucoside decreased by287.50%.And compared with HangbaiJu, it had decreased by262.50%. And compared with ChuJu, it had decreased by12.50%.
