| In order to form the preliminary DUS test guidelines,31varieties of woad seeds collected from domestic main producing areas were cultivated in the field and then the botanical and quality characters were investigated and studied. Based on the research of DUS test guidelines, several main types of I. indigotica Fort, were screened as the experiment materials to study the salt tolerance and waterlogging resistance. Concrete contents were as follows:1. One qualitative binary character,12qualitative polymorphic characters and16quantitative characters were observed in the field. The distinctness, uniformity and stability of those characters were analyzed by using coefficient of variation, T-test. The result showed that the uniformity of all qualitative binary characters and most qualitative polymorphic characters were good for all samples, while those quantitative characters were influenced by the environment significantly. The stability of qualitative binary characters were best and qualitative polymorphic characters were relatively good. However, the stability of quantitative characters varied most. On the basis of analysis,22pre-requisite characters and6optional characters were screened for the DUS testing.2. Main active components in both leaf and root of the woad materials were investigated. The results showed that contents of active components have great difference among the samples. The content of indigo was between1.64~10.18mg·g-1, content of indigo in SL-29was highest which was6.21times of that in SL-04. The content of indirubin was between0.39~3.22mg·g-1, content of indirubin in SL-23was highest which was7.36times higher than that in SL-30. The content of R,S-goitrin in19samples was between0.60~3.50mg·g-1,73%of all samples. The content of polysaccharide was between4.00%~8.50%, the average value was6.10%.3.I. indigotica Fort, of chinese cabbage leaf type, cabbage leaf type, mustard leaf type and tetraploid were chosen as experiment materials to be pot-cultured, and then the seedlings were treated with50mmol·L-1NaCl, group that treated with distilled water was set as the control. The results showed that the salt treatment significantly decreased the fresh weight and dry weight of every single plant; as the salt treatment went by, the value of Pn, Tr, Gs, SPAD all showd to be obviously decreased, while the value of Ci was decreased at first and then increased; content of chlorophyll was significant lower compared to the control group, the ratio of ch1a to ch1b showed a decreased trend for that the content of ch1a decreased faster than ch1b; content of R,S-goitrin in the root decreased under the salt stress, while polysaccharide content was significantly increased.4.I. indigotica Fort, of chinese cabbage leaf type, cabbage leaf type, mustard leaf type and tetraploid were chosen as experiment materials to study the effects of mild waterlogging stress, moderate waterlogging stress and severe waterlogging stress on physiological and biological characteristic of the seedlings, and then comprehensive evaluation of waterlogging resistance of the four types of I. indigotica Fort, was made by using principal component analysis. The results showed that with waterlogging stress become more and more serious, the ratio of injured leaf area, relative conductivity, content of MDA increased significantly; content of total ch1decreased gradually; activity of antioxidant enzyme increased as well as the activity of ADH in the root; content of osmoregulation substance such as proline and soluble sugar increased significantly; the comprehensive evaluation result showed that the waterlogging resistance in series from strong to the weak was:mustard leaf type>cabbage leaf type>tetraploid>chinese cabbage leaf type.5.I. indigotica Fort, of chinese cabbage leaf type was chosen as experiment material and a pot experiment was conducted. Related enzymes activity and physiological indicators of woad treated by different concentrations of EBR (0.01ã€0.05ã€0.10ã€0.50ã€1.00mg-L"1) were investigated. The results showed:When the concentration of EBR were0.10,0.50mg·L-1, the woad had the least number of yellow leaves; with the increase of concentrations of EBR, antioxidant enzyme, ADH, NR activity increased at first and then decreased, as well as the trend of Pro and soluble protein contents, compared to the opposite change of MDA. |
