Collagen is a kind of white opacity native celloglobulin without branched chain, and it isresided in skin, skeleton, teeth, tendon and ligaments, blood vessel generally. collagen is themost profuse content in the mammal body, the main function of which is to keep theappearance and construction of skin and organs, in order to support organs and protectorganism. Collagen is special nature biotic resources with favourable trophic function, andphysiological function, so it is applied in the medicine, foodstuff, cosmetics and tanningextremely widespread. It is generally proved that collagen active peptide from proteindegradation is better for human body, and get nearly the same function with collagen.In this paper, first the waste cod skin was used as raw material to extract collagen,offering N source to take the place of NaNO3for black mold, mold sp.Rapid, mold sp.3800,mold sp.3042, mold sp.39screening strain in order to get the strain with collagenase anddetermine the enzyme activity by ninhydrin colouration. Then, we put extracted collagen andcollagenase to carry out enzyme reaction in order to determine the optimum condition. Finally,we analyze collagen protein active peptide by paper chromatography and SDS-PAGEelectrophoresis method.The results showed that both black mold and mold sp.Rapid had better collagenaseactivity than the others, and their collagenase activity were35.97U,30.08U respectively. Theoptimal enzyme reaction condition were as follows: the reaction time was20h, temperaturewas42℃,pH was5.5. The products of enzymolysis were major of collagen activepeptide,and the weight of it was about4kDa.
