Extract from purple sweet potato is an efficient antioxidant, which contains many kinds of chemical composition with physiological activation. This paper studied purple sweet potato extraction’s function of anti-aging and explored its body oxidation resistance with the modle of Drosophila melanogaster.Purple sweet potato anthocyanin which is the major active component of the sample had been quantitated by MS. The column was ODS column (250mm×4.6mm,5μm); peak spectra was520nm. A binary H2O-0.1%CH2O2(solvent A) and acetonitrile (solvent B) gradients was used for elution. It was showed that compositions were the acylation of peonidin (cyanidin)-3-sophoroside-5-glucoside.Lifspan test and biochemical analysis methods were used to discuss the effect of extract from purple sweet potato on the lifespan and antioxidant system of Drosophila. Every test groups prolonged the lifespan of Drosophila, with the increase of extract from purple sweet potato; aging was delayed in a dose-response relationship.16mg/mL of purple sweet potato extraction was the most effective dose with45days compared with the blank value (38days), increasing by18.4%.Lifspan test and biochemical analysis methods were used to discuss the protected effect and the mechanism of extract from purple sweet potato about the damage of lipid per-oxidation result by used10%lard in fruit flies and by UV. The present study found that dietary fat and UV accelerates the aging process and causes a greater mortality by accumulating lipid hydroperoxide in fruit flies. The mean lifespans of flies fed on16mg/mL of purple sweet potato extraction were prolonged44.89%、23.4%, respectively. Total SOD, CuZn-SOD, CAT enzyme activities were improved and the content of MDA reduced in a dose-response relationship.Hydrogen peroxide treatment and paraquat treatment were used to exmine the resistance of flies agaist oxidantive stress. The study revealed that supplementation of16mg/mL extract from purple sweet potato prolonged the survival time of fruit flies.The real-time fluorescence quantitative PCR experiments on SOD1、SOD2、CAT mRNA of flies showed as follows. Compared with the blank group, SOD1、SOD2、CAT gene expression level was significantly increased.The results of experiments showed that extract from purple sweet potato possess strong antioxidant activity and anti-aging activity.
