Microbial enhanced oil recovery technology (MEOR) use of microbe activities andmetabolites (organic acids, gases, surfactants, polymers.etc) which effect on reservoir fluidand reservoir rock internal pore surface to improve mobility of crude oil and oil-waterinterfacial condition. MEOR is known as the fourth important EOR technology aftertraditional EOR technologies such as thermal driving, chemical flooding and gas flooding,Compared with other EOR techniques methods, MEOR have wide application scope, simpleprocess, low investment, low cost, no damage to the reservoir and non-pollutingcharacteristics. Chang6oil-bearing reservoir in HuaZiping area of Ansai Oilfield have poorphysical properties, low pressure, low natural energy and ow recovery percent, it is difficult touse traditional recovery technology, MEOR support new ideas and and technologies forfurther exploration.This paper is study on the method of gradient dilution coating and plate streaking,separated and purified14bacterial strains and11fungal strains from crude oil,formationwater and sludge in Hua208well, Huaziping oil area. After primary screening and secondaryscreening, pick one fungal strain and one facultative anaerobe strain, then evaluate the twostrains’ reservoir adaptability to ensure that bacteria in the reservoir can be growing well.Gathering the bacteria which can adapt to the reservoir environment and using crude oil assole carbon source, then mixed with the crude oil. Analysis of the crude oil physicalproperties (adhesion, viscosity) and determined the change of crude oil composition by themethod of gas chromatography. Using clastic rock thin sections to observe the scavengingeffect of crude oil in the microscopic pore channels.The experimental results showed that: after screening, most of the bacteria can grow wellon the plate, but on the inorganic salt medium the bacteria which using crude oil as the solocarbon source have great difference in growth, in these bacteria GC3and G1grow well. In thesubsequent evaluation, two species can adapt to the reservoir environment, and to some extent reduces the oil viscosity, can decrease the pH value of the culture solution, After thechromatographic analysis of treated crude oil, the light component was increases.By16SrDNA sequencing techniques to determine GC3strain is Bacillus tequilensis,homology was99.93%. This paper aimed to screening efficient oil recovery bacterial inChang6oil-bearing reservoir from Huaziping oil area, Ansai Oilfield. Finally give themtheoretical and technical support for future applications in oil field.
