Threonine is mainly produced by fermentation. In its process, a large amount of second mother liquor of threonine is produced, which contains pigment, sugar, inorganic salt, threonine and protein, and the contents of threonine is relatively high. If these mother liquor is directly put out into environment or sold for feed stuff additive factory, not only the value of them fall, but also they make large pollution. At the same time, domestic and international market have a steady growth demand for threonine, which is one of the fastest varieties of amino acids, so it has great economic benefits for recycling threonine. In this study, the second mother liquor of threonine was taken as the main materials which was from EPPEN. The second mother liquor was pretreated first, and then put into the chromatographic separation. At the same time, the regeneration of macroporous adsorption was did, finally the new and more effectively ways were gained, which could provide some reference basis for its industrial application. The main results are as follows:(1)The contents of amino acid, total sugar and inorganic salt in the mother liquor accounted for16.12%,4.66%and7.70%, respectively.(2)The technology for separation and recovery threonine using bed type was studied and concluded that a better technological conditions:the feed temperature was55℃, the feed flow rate was0.74BV/h, the elution temperature was55℃, the elution velocity was0.74BV/h, the recovery rate of threonine reached84.19%.(3)The technology for separation and recovery threonine using chromatography coupling ultrafiltration was studied and concluded that a better technological conditions:the feed temperature was50℃. the feed flow rate was0.74BV/h, the elution temperature was50℃, the elution velocity was0.74BV/h, the recovery rate of threonine reached95.26%.(4)The regeneration technology for CT-101macroporous adsorption resin using static experiments and dynamic experiments was studied. Through static experiment, it concluded that the better regeneration agent is alkaline ethanol, and the better technology conditions as follows:the concentration of NaOH was4%, the concentration of ethanol was65%and the ratio of alkali alcohol was3:7, through dynamic experiment, it concluded that the better regeneration process conditions as follows:the velocity was1.50BV/h, the elution liquid product was3.50BV and the operating temperature was30℃.
