Study On Hydrolysates Of Duck Embryo Egg In Promoting Wound Healing

Embryo egg has been recognized as a perfect supplement and there is a folk habit of eating Embryo egg earlier, but there is rare research about its nutrition composition and functional properties, let alone the market supply of processed product. Given the fact there are a great number of dead embryos besides living embryos during the period of poultry incubation, and there are a series of complicated biochemical process in the process of incubation, which may produce active substances such as peptides that contribute to the growth of organism. To take advantage of this natural and complex biological transformation process, in this study embryo egg was first digested by enzyme and then the possibility that hydrolysates of duck embryo egg may promote wound healing and the relative molecular or cellular mechanisms were illustrated through biochemical and cell test. This study provide a reference for developing cosmetic ingredients using eggs as raw material, provide an initial foundation for further purification and animal experiments and expand its application in food, medicine, cosmetics field. The main contents and results are as follows:(1) The contents of Ca, Mg, Zn, Cu and other elements increased with extension of incubation and the content of total protein, soluble protein, free amino acids and peptides were higher in late incubation period than that in the initial incubation period; Simulated human digestive test showed that the amino acids, peptides significantly increased, and the peptides of F sample were significantly higher than that of other groups.(2) Hydrolysates of duck embryo egg in six different hatching states can promote the proliferation of epidermal cells HaCaT, and the repairing ability and the cell proliferation ability showed a good dependence on duck embryo egg F hydrolysates when the concentration was within12.5μg/mL~100μg/mL. Cell migration test results showed that the cell proliferation played a dominant role in promoting wound healing. Hydrolysates of duck embryo egg could significantly improve the expression levels of EGF and IGF1, inflammatory cytokines TNF-a, IL-6and IL-10, but has no significant effect on FGF2.(3) Hydrolysates of duck embryo egg in six different hatching states can promote the proliferation of fibroblasts HSF, and the repairing ability and the cell proliferation ability showed a good dependence on duck embryo egg F hydrolysates when the concentration was within12.5μg/mL~100μg/mL. Cell migration test results showed that the cell proliferation played a dominant role in promoting wound healing. Hydrolysates of duck embryo egg significantly improved the expression levels of EGF、IGF1、FGF2, PDGFA and inflammatory cytokines TNF-a, IL-6and IL-10.(4) Hydrolysates of six kinds of duck embryo egg in different hatching states could promote the proliferation of macrophages RAW264.7when the concentration was within a certain range. Meanwhile, hydrolysates of duck embryo egg F, in the range of12.5μg/mL~100μg/mL, can significantly enhance the phagocytosis ability for neutral red and releasing ability of NO of macrophages and it showed a concentration denpendence. It can be initially explained that duck embryo egg hydrolysates can accelerate wound healing through promoting proliferation of macrophages and activating its vitality.
