In this paper, small black beans whey protein was obtained from small black beans, it was separated and purified by membrane separation technology. Whey poly-peptides were prepared with single enzyme and many enzymes hydrolysis, the best anti-tumor effect was selected by S180and H22tumor cells in vitro. The study was arounded by immune function,anti-S180-tumor, anti-Skin papilloma and chemical resistance of liver cancer in mice of whey poly-peptides and whey protein.The results was as follow:1. Black soybean milk water was pretreatmented with flocculation centrifugation, the optimum technological conditions for:pH8.0, CaCl215ml·L-1,40℃. Then small black beans whey protein was obtained through microfiltration and ultrafiltration. The results showed by Sephadex G-50gel analysis was that molecular weight range of small black bean whey protein was in6.0KD-10.0KD, which accounted for50.3%, macromolecular which was higher than10.0KD accounted for about45%.2. The comparative analysis of small black beans and yellow soybeans showed that, whey protein, ash content and total sugar content in small black bean milk were higher than those in soybean whey water; Electrophoretic analysis showed that molecular weight of small black beans whey protein and soy whey protein were mainly distributed in below30.0KD. BBI content in small black beans was higher than soybean whey protein.3. Small black bean whey protein was hydrolyzed with a single protease and double compound enzyme. The single bacterial acid protease enzyme reached the highest extraction rate of peptide, which up to97%.The best double compound enzyme was pepsin-bacterial acid protease, the rate was63.36%. Sephadex G-25gel analysis showed that the polypeptide which was less than1kd accounted for more than80%.4. Many kinds of peptides in vitro antitumor experiment were carried out in this paper. The conclusions were as follows:the effect of peptide hydrolyzed by papain was best on S180and H22antitumor. Against the best antitumor effect, orthogonal experiment of peptide hydrolyzed by papain was designed, optimal polypeptide preparation technology was enzyme amount5000u/g protein, pH6.0, processing temperature65°C.5. Immune function of small black beans whey protein and polypeptide mice was carried out in this study. It was found that there was no obvious inhibitory effect on the mice immune organs. At the same dose, the75℃treatment group was significantly better than the treatment of65℃group to inhibit the action of various immune function decline, the concentration-response relationship existed to some degree. The effect of enhance the immune of peptides was superior to the protein group, but polypeptides dose negatively related with antitumor effect.6. Resistance against S180tumour cell, skin papilloma and precancerous lesion of liver cancer was learned, mice were fed with whey protein and polypeptide. Experiment showed that antitumor activity of75℃high dose group was better than65℃protein group. its tumor inhibition rate amounted to41.8%. The preferably resistance against skin papilloma was proteins and peptides with high dose group, tumor suppression effect was positively correlated to concentration. The resistance of liver cancer results showed that the effect of inhibition of liver cancer cell growth of Prote AX peptide group was superior to the papaya polypeptide group, the anticancer effect was positively correlated to peptide concentrations. Therefor, small black beans whey protein and peptides played a role in preventing tumors.
