Objective:Atherosclerosis is the major cause of death from cardiovascular and cerebrovascular diseades. We intend to investigate the effect of serum containing Huanglian Jiedu decoction (HLJDT) on regulating the phenotype of macrophages and foam cells and provide experimental evidences for future clinical use.Materials and Methods:1. Bone marrow-derived macrophages (BMDMs) and foam cells were primarily cultured from bone marrow cells of C57mice, and detected by Hematoxylin and Oil Red O staining technique.2. Serum containing HLJDT was derived from rats given HLJDT gastric gavage.3. Bone marrow cells were stimulated by M-CSF for7days to differentiate BMDMs with or without serum containing HLJDT; BMDMs were stimulated with or without serum containing HLJDT for another3days; BMDMs were stimulated by ox-LDL to form foam cells with or without serum containing HLJDT; foam cells were still stimulated by ox-LDL with or without serum containing HLJDT. The subtypes of macrophages and foam cells were detected by FACS. the genes of Nos2and Argl in these cells were assayed by Real-time PCR.Results:1. Scrum containing HLJDT induced bone marrow cells to differentiate to M2macrophages by increasing the expression of CD206and Argl and decreasing the expression of Nos2.2. Serum containing HLJDT further induced BMDMs to differentiate to M2subtype.3. For short-term stimulation, ox-LDL induced BMDMs to differentiate to M2subtype of foam cells, and serum containing HLJDT increased the differentiation of BMDMs to M2subtype of foam cells.4. For long-term stimulation, ox-LDL induced M2subtype of foam cells to differentiate to M1subtype, and serum containing HLJDT inhibited M2subtype of foam cells to differentiate to M1subtype under long-term ox-LDL and LPS/IFN-γ inflammatory stimulation. Conclusion:In vitro, serum containing HLJDT can inhibit atherosclerosis by regulating differentiation of macrophages and foam cells.
