Objective:This study based on the solublization function of glycyrrhizin in Gancao(Radix Glycyrrhizae).We researched all the prescriptions that contains both Radix Glycyrrhizae and the other herbs whose active ingredient are indissolvable, and the prescription should has fewer compounds which is easy to study. At last, gegenqinliantang was selected which includes Radix Puerariae Lobatae and Radix Scutellariae Baicalensis. Puerarin in Radix Puerariae Lobatae and Baicalin in Radix Scutellariae Baicalensis are the indissolvable ingredients. So the research aim to find out the solublization function of glycyrrhizin to Puerarin and Baicalin, also to summarize the regularities and mechanism, so to provide references to the solublization of indissolvable ingredients in Chinese drugs preparation.Methods:1. Establish the assaying methods of Puerarin, Baicalin and glycyrrhizic acid.2. Research on the critical micelle concentration of glycyrrhizin and its influencing factors.3. Establish the characteristic fingerprint map of the prescription according to its compatibility percentage and the solublization function of glycyrrhizin.4. Effect of pH in best compatibility of Radix Puerariae and Radix Glycyrrhizae.Results:1. After investigating on extraction solvents and methods, we decided the best extraction methods of these three herbs.30%methanol, ultrasound0.5h was the best for Radix Puerariae,70%ethanol, circumfluence,3h for Radix Scutellariae Baicalensis, and70%ethanol, ultrasound0.5h after soaking12h for Radix Glycyrrhizae. Simultaneously determination of the target ingredients of them was established.Research methodology study was qualified and determination results were accurate.2.From the point of view of traditional boiling method to investigated the compatibility of Radix Glycyrrhizae with Radix Puerariae, Radix Glycyrrhizae with Radix Scutellariae Baicalensis, we found out that when the ratio of Radix Puerariae to Radix Glycyrrhizae was3:1, extraction rate of Puerarin was maximized, and Radix Scutellariae Baicalensis to Radix Glycyrrhizae was6:1, best for Baicalin. From the point of view of the physical and chemical effects of Radix Puerariae and Radix Glycyrrhizae, Radix Glycyrrhizae with Radix Scutellariae Baicalensis, We found the ratios mentioned above were changed to5:3and3:2.3. According to the studies solublization function of glycyrrhizic acid to Puerarin and Baicalin, we established the fingerprint according to the original parties of gengenqinliantang without Rhizoma Coptidis, finding no new substances afte compatibility of, only the changes of contents.Conclusion:1. Glycyrrhizic acid in Radix Glycyrrhizae has solublition function. When reaching its critical micelle concentration(CMC), it can form supermolecule parceling indissolvable ingredients like Puerarin and Baicalin, so to increase their solubility and improve bioavailability. The CMC of glycyrrhizic acid is0.2mg/ml. When accompany with Radix Puerariae Lobatae according to their best compatibility, its CMC changed to0.18mg/ml, and Puerarin reached its biggest dissolubility.CMC was0.22mg/ml when with Radix Scutellariae Baicalensis according to their best compatibility, Baicalin reached its biggest dissolubility. So this powerfully testified other members’result using computer simulating, the CMC of which is0.2mg/ml, and varied with different solubilization system. Meanwhile we conclude that CMC of glycyrrhizic acid misgrated because of the exist of Baicalin and Puerarin, but the misgration was not that big. The CMC was related to the structure of the ingredients.2. Radix Puerariae Lobatae,Radix Scutellariae Baicalensis and Radix Glycyrrhizae, decocting alone,both, or together had different dissolution rate, which was related to mul-compatibility system that making for forming CMC, so to play its solublization effect.3. Although we testified that the three of them decocting together was benefit for forming CMC and improving dissolubility, there is still some insufficiency needed deep investigation.4.we established the fingerprint according to the original parties finding no new substances afte compatibility of, only the changes of contents.5.pH played a role in forming CMC of glycyrrhizin, and then affected best compatibility of Radix Glycyrrhizae and Radix Puerariae Lobatae. |
