| ObjectiveTo study streptozotocin （STZ）-induced type2diabetic rats iris pathology （theneovascularization of iris of diabetic, etc.）, is to unveil the causes of the pathologicalchanges in the iris of the type2diabetic rats. Meanwhile，it discussed the relationshipbetween the formation of the new vascularization of diabetic rat iris and the quantity ofthe CTGF existed in iris and the relativity between them.Method60Wister rats were randomly divided into experimental and control（negativecontrol group and positive control） groups. Every group was20rats.1． The experimental group was prepared by intraperitoneal injection of STZ to get theiris neovascularization and fibrosis rat model of type2diabetes.2. The positive controlgroup was prepared by intraperitoneal injection of STZ to not get the irisneovascularization and fibrosis rat model of type2diabetes. The negative control groupwas normal.3. After the succession of modeling， the106rats’ eyes were taken out. Theexperimental group and control group of15eyes were used enzyme-linkedimmunosorbent assay for quantitative analysis of the content of CTGF in iris. Anotherexperimental group of13eyes, positive control group of23eyes and negative controlgroup of25eyes were taken the iris for light and electron microscopy (Electronmicroscope-10eyes of experimental group and control group, another eyes wereobservation by the light microscopy).Result1. Clinical signs：Negative control group rats form was normal,and those eyes nosignificant change;When the compound tropicamide eye drops one drop was put intothese eyes, Pupil was dilated after ten minutes. The form of positive control group ratswere thin,and these drank a lot of water,but the eye had no significant change. When thecompound tropicamide eye drops one drop was put into these eyes, Pupil was dilatedafter ten minutes. The experimental group rats’ form were thin,and around the irisappeared the neovascularization. When the compound tropicamide eye drops one dropwas put into these eyes, Pupil was not dilated after ten minutes.2． Compared with the experimental group and control group, experimental group hadsignificantly NVI iris characteristic changes and appeared fibrosis but the control grouphad no significant change under the light and electron microscopy.3. Average value of CTGF was1010.49ng·L-1of the iris in experimented group,meanwhile, it was644.82ng·L-1of the iris in negative control group, was749.66ng·L-1of the iris in positive control group.The purity of CTGF of the iris in the experimentalgroup, which had a significant difference compared with the control group’s （P<0.001）.ConclusionThe purity of the CTGF of iris is positively correlated to the generation of new vascularization of iris and fibrosis of iris in the type2diabetic rats. |
