Objective:Measured the expression of Phosphorylated Protein Kinase B(pAKt) of Uterus Homogenate by Western Blotting Assay in mouse peri-implantation receiving controlled ovarian hyperstimulation, and test the content of nitric oxide synthetasev(NOS) of Mouse serum by enzyme-linked immunosorbent assay (ELISA) to explore the effect and its possible mechanism of Complex Kunyuan decoction to mouse uterine microenvironment on Embryo Implantation,in order to provide theoretical basis for the application of traditional Chinese medicine in embryo implantation.Methods:All Kunming mice are randomly classified into five groups: blank control group, model group, Kunyuan decoction in low concentration group, medium concentration group and high concentration group.The mice will be killed in fifth days after establishment of mice model of embryo implantation,and then test the content of NOS of Mouse serum by enzyme-linked immunosorbent assay and the expression of pAKt of Uterus Homogenate by Western Blotting Assay.Results:The number of mouse blastocyst receiving controlled ovarian hyperstimulation was significantly higher than blank control group(P 0.05),but the expression of Phosphorylated Protein Kinase B was lower than blank control group.The expression level of PAKt in traditional chinese medicine intervention group increased significantly,and it’s not statistical significance that Kunyuan decoction in medium concentration group close to blank control group (P>0.05)②The content of NOS of model group was significantly higher than blank control group and Kunyuan decoction groups(P<0.01), and the blank control group was significantly higher than Kunyuan decoction in low concentration group and high concentration group(P<0.01),at the same time it’s not statistical significance to compare with Kunyuan decoction in medium concentration group.The content of NOS of Kunyuan decoction in high, low and medium concentration group successively rised,and there were statistical significance(P<0.05, P<0.01).Conclusion:①The tcontrolled ovarian hyperstimulation could change the content of NOS and expression of Phosphorylated Protein Kinase B,and Change the uterus local microenvironment,thus restrained the occurrence of embryo implantation.②Traditional Chinese medicine Kunyuan decoction improved the uterus local microenvironment by measuring the content of NOS and expression of pAKt, so as to achieve the purpose of promoting embryo implantation in mice.
