Drug addiction is a chronically-relapsing brain disease that characterized by losing control ability and continuous compulsive drug use, it caused by the combined effect of biological, behavioral, and environmental factors. Long-lasting vulnerability to relapse has been recognized as a phenomenon pivotal for the understanding and treatment of drug addiction. It create a recreational effect at the initial stage of drug use, and the adaption of central nervous system will change due to chronic use. Withdrawal symptom occur when abstinence. The drug self administration and extinction-reinstatement procedure is a acknowledged ethological model of relapse of drug addiction that, effectively model the diverse aspects of the addiction cycle, such as intravenous self administration behavior, drug extinction, drug triggered reinstatement, drug-related environment stimulus cued reinstatement and stress induced reinstatement. The great majority of self administration and extinction-reinstatement procedure use simple fixed ratio1(FR1) schedule, which is useful for exploring patterns of rate of drug intake, and the procedure of’the model is:model the self administration behavior, then extinct the behavior, and at last, process the reinstatement test.The present research aimed at:the establishment of rat cocaine self administration and extinction-reinstatement procedure, and discuss the modeling procedure; then the influence to the cocaine triggered reinstatement by Tat-A2which, a synthetic LTP blocking peptide will be measured and discussed.The major results were described as follows:1. Basic and critical processes to establish the self-administration model are implantation and maintainence of indwelling jugular catheters. Otherwise, operating process, catheter size, catheter patency, and infection or inflammation all effect the duration of the catheter..2. Establishment of rat cocaine FR1self administration behavior. After a6days researcher assisted training phase and a14days independent training phase, the average injection of cocaine (0.75mg/kg/inj.) developed and stabilized at33.17±1.45in2hours, the unactive nosepoke decreased with the processing of the session. 3. Establishment of rat cocaine FR1self administration extinction session. Cocaine injection was replaced by saline in this session, and the cocaine-related environment stimulus cues (light and tone) were maintained. There are20rats finished this session and reached the criterion of extinction as well, after a10days extinction phase, the average injection of saline decreased and stabilized at3.86±0.50in hour, the unactive nosepoke decreased with the processing of the session.4. Synthetic Tat-A2has no significant effects on cocaine triggered reinstatement. |
