In this article, Polysaccharides were extracted and purified from the Lethariella cashmeriana.ThePhysicochemical properties、structure and non-alcoholic fatty liver disease (NAFLD) preventive effect has been systematically studied, The results showed as following:The best water extraction of the liquid material18.73mL/g,1.1h of the extraction time, extraction temperature of128℃. The best alcohol precipitation process for the liquid material11.10mL/g, and alcohol precipitation time36.42h,77.57%alcohol content. Best addition to protein technology for the enzyme1.5U/mg, hydrolysis time was1h, hydrolysis temperature was50℃, the initial pH of5. In this process of crude polysaccharide (LP) was9.9%, which contains polysaccharide89.0%, protein content0.35%.During the purification of crude Polysaccharides (LP), two polysaccharides were isolated by using DE-52and Sephadex G-100, they were named LPP1and LPP2.The uronic acid content of LP and LPP2were16.4%,34.1%. LP contain five components by GPC analysis. They were named Ⅰ1,Ⅰ2,Ⅰ3,Ⅰ4,Ⅰ5. The weight average molecular weight (Mw) of LP order664365,256986, 91324,25377,7483. LPP1, LPP2showed a narrow single symmetrical peak, both Mw354703,40117. The experiment shows that the LP contain D-glucuronic acid, D-glucose, D-mannose, D-xylose, fructose,(1:3.125:1.107:2.212:1.751), LPP1containing D-glucose, D-xylose, fructose (3.128:1:3.362), LPP2containing D-glucuronic acid, D-glucose, D-mannose (2.098:3.277:1).High-fat and high-sugar diet can cause the rat NAFLD model. Polysaccharide of Lethariella cashmeriana can reduce the rat liver wet weight and liver index, improve lipid parameters (TC, TG, LDL-C HDL-C), increase the rat of SOD and GSH activity, inhibition of lipid peroxidation the increase of MDA content, and enhance the role of the antioxidant capacity in rats, prevent and treat NAFLD effectivily.
