Cold Blood Detoxifying Decoction Xierexing Curative Effect Observation With Psoriasis Vulgaris And The Influence Of Related Factors On The Th17 Cells

Objective:The study is to evaluate the safety and effectiveness of Liangxue Jiedu Decoction, and investigate the treatment mechanism of Chinese herbal compound prescription by the research of Liangxue Jiedu Decoction efficacy for patients with psoriasis vulgaris of blood-heat type, and by influence of Liangxue Jiedu Decoction On the expressions of Th17cells related cytokines IL-17A, IL-17F, IL-21, IL-22,and transcription factor RORC2and cells surface marks IL-23R in PBMC of patients with psoriasis vulgaris of blood-heat type.Methods:According to the selected standard,we collected30patients with psoriasis vulgaris of blood-heat type and20healthy controls, whose PBMC (peripheral blood monouclear cell) were drawn.After6weeks treatment by Liangxue Jiedu Decoction,the same quantity PBMC were drawn from those patients, then we draw RNA from PBMC for Real-time RT-PCR experiments. The effectiveness of Liangxue Jiedu Decoction was evaluated by PAS1(psoriasis area and severity index) and QOL (quality of life) scores,and the area of psoriatic lesions, The safety of Liangxue Jiedu Decoction treatment was evaluated by tests of blood and urine routine, liver function and renal function.Targets of Liangxue Jiedu Decoction treatment was analysed by relative expression quantity of RORC2JL-17A, IL-17F, IL-21, IL-22and IL-23R mRNA in PBMC of patients before and after the treatment. Pathogenesis of psoriasis was investigated by relative expression quantity of RORC2and1L-23R mRNA in PBMC of patient group and contol group.PASI, QOL and the area of psoriatic lesions were Analyz by rank-sum testRelative expression quantity of RORC2JL-17A, IL-17F, IL-21, IL-22. IL-23RmRNA in patiens gruop and control group were Analyz by t-test.Results:1The effective rate of Liangxue Jiedu Decoction treatment efficacy for psoriasis vulgaris of blood-heat type93.3percentage.2After the treatment of Liangxue Jiedu Decoction, the grade of PASI and QOL, the area of psoriatic lesions were found to be significantly ameliorated (P<0.05).3Levels of RORC2,IL-23RmRNA were found to be significantly higher in all psoriatic PBMC compared with that of normal controls (P<0.05).4After the treatment with Liangxue Jiedu Decoction,the levels of RORC2,IL-17A,IL-17F,IL-21.IL-22and IL-23R mRNA were found to be lower.the Relative expression quantity of RORC2,IL-17A mRNA, IL-17F,IL-21, IL-22,IL-23R mRNA is0.41、0.31、0.2、0.35、0.9、0.36、0.31times compare to that before.Conclusion:1Liangxue Jiedu Decoction is effective and safe in treatment of psoriasis vulgaris with blood-heat type.2The treatment effectiveness might work by down-regulating the level of RORC2, IL-17A,IL-17F,IL-21, IL-22and,23R mRNA.3RORC2and IL-23R might be related to the activation and proliferation of Th17cell, Which might be involved in the pathogenesis of psoriasis.
