| Objective:To investigate the single-nucleotide polymorphism(SNP) at position rs734553of intron7of SLC2A9gene for the association with hyperuricemia (HUA) with diabetes mellitus(DM) in the Chinese Han population in Qingdao City of Shandong province.Methods:A segment of intron7of SLC2A9gene was amplified by sequence specific primers and polymerase chain reaction (PCR) method, then all PCR products were digested by restriction enzyme HapII and a part of PCR products were submitted to DNA sequence analysis, and we detected the differences of the allele frequency and genotype frequency of SLC2A9gene at position rs734553in709patients in affiliated hospital of medical college of Qingdao university in Shandong province.Results:A allele frequency of rs734553polymorphism of intron7of SLC2A9gene in control group is97.75%, in DM group is99.04%, in HUA group is99.07%, and in HUA with DM group is99.5%, while C allele frequency in control group is2.25%, in IGT/IFG group is0.96%, in HUA group is0.93%, and in HUA with DM group is0.50%. The difference of allele frequency between control group and HUA with DM group had statistical significance CP<0.050), the difference of allele frequency in other groups had no statistical significance (P>0.05).The AA genotype frequency in control group is95.5%, in DM group is98.1%, in HUA group is98.1%, and in HUA with DM group is98.9%, the AC genotype frequency in control group is4.5%, in DM group is1.9%, in HUA group is1.9%, and in HUA with DM group is1.1%. The genotype frequency in those groups had no statistical significance (P>0.05).Conclusion:In the Chinese Han population in Qingdao City of Shandong province, rs734553polymorphism of intron7of SLC2A9gene is assicated with HUA with DM, the incidence of HUA with DM in those people with C allele is higher than those with A allele. |
