Objective:To investigate the intervention effect of compound Yindan decoction in rats with acute intrahepatic cholestasis induced by alpha-naphthyl isothiocyanate (ANIT).to observe the effect of Traditional Chinese Medicine extract on the general condition,hepatic function and hepatic tissue pathological changes of rats with intrahepatic cholestasis.Methods:45male wistar rats were randomly divided into three groups, the model group (18),TCM group (18),normal control group (9),according to the different processing time,each group is divided into24-hour,48-hour,72-hour three sub-groups in turn, in which model group,TCM group for each sub-group of6rats,and normal control group for each sub-group of3rats were raised separately.The rats of model group,TCM group and normal control group were given separately2%ANIT and volume of salad oil by gavage with once administrat ion. After2hours of modeling,the rats of TCM group and model group and normal control group were given separately lml/100g weight of TCM and volume of saline by gavage once a day. Observating dynamically the general condition,hepatic function and hepatic tissue pathological changes of rats in24-hour,48-hour,72-hour three time points of modeling.Results:1General condition:Comparing with normal control group,the hairs of the rats of model group was scattered and had no luster, their activity values and eating were deficiency,the urine of which was dark yellow, their weight decreased in varying degrees, decreasing significantly at modeling48hours and72hours(P<0.01); liver weight and liver index dynamic increased correspondingly(P<0.01). Comparing with model group,the activity values,eat ing and the color of urine of the rats of TCM group were improved,and their weight significantly increased and liver weight and liver index significantly decreased at modeling72hours (P<0.05).2Hepatic funct ion:ALT, AST. TB i1. DBil, GGT, ALP and TBA of model group were significantly increased compared with normal control group (P<0.01),ALT,AST.TBil.DBil and ALP reached the peak at modeling48hours,GGT reached the peak at modeling72hours,TBA reached the peak at model ing24hours. ALT,AST,TBi DBil,GGT. ALP and TBA of TCM group were decreased with the administration time extension,and all biochemical indexes were significantly decreased at72hours compared with model group(P<0.05,P<0.01).3HE staining of hepatic tissue pathological changes:Comparing with normal control group,the rats of model group appeared damage to the hepatic lobules structure,hepatocyte swelling,and scattering and exfoliation of biliary epithelial cells and infiltration of neutrophils in portal area at24hours after modeling; hepatic tissue lesions aggravated,hepatic lobules structure destructed severely,nucleus of different sizes, nuclear chromatin gathered,nucleolus enlarged and hyperstained. At48hours,we can see that significant hyperplasia of biliary epithelial cells,punctate and focal necrosis of hepatic cells around biliary and a lot of infiltration of neutrophils in portal area; At72hours.it can be still seen the hyperplasia of biliary epithelial cells and infiltration of neutrophils in portal area.The hepatic tissue pathological changes of TCM group were not very significant at24hours and48hours compared with model group,but neutrophils infiltration significantly decreased at72hours in portal area.Conclusions:1In this study,we replicated the model successfully of acute intrahepatic cholestasis by ANIT which was administered by gavage.2The experimental results confirmed that compound Yindan decoction can improve the general condition of intrahepatic cholestasis in rats, decrease serum bilirubin level, transaminase and duct enzyme,protect hepatic function,reduce hepatocellular swelling and capillary bile duct inflammation and improve hepatic histological lesions and has a certain effect on intrahepatic cholestasis induced by ANIT.
