| Objective To observe the inhibition effect of Methylprednisolone on cell human glomerular mesangial cell in vitro, and explore its mechanisms by measuring the expression of cycle proteins and CKD kinase.Methods (1) MTT method was used to investigate the inhibition effect of different concentrations of Methylprednisolone (1mg/Lã€10mg/Lã€100mg/L) on Human glomerular mesangial cell proliferation.(2) Flow Cytometric method was used to detect the proteins expression of CyclinE, CyclinD1, P27Kip1, CDK2and CDK4in Human glomerular mesangial cell which was handled by different concentrations of Methylprednisolone (1mg/Lã€10mg/Lã€1OOmg/L).(3) Western blot analysis was used to identify the expression of CyclinE, CyclinD1, and P27Kip1in Human glomerular mesangial cell which was handled by different concentrations of Methylprednisolone (1mg/Lã€10mg/Lã€100mg/L)Results (1) Compared with the control group, Methylprednisolone inhibited human glomerular mesangial cell proliferation in a concentration depend measure in the concentration of1-100mg/L.(2) After Flow cytometric test, compared with the control group, Methylprednisolone role in human glomerular mesangial cells, the protein expression of CyclinE, CyclinD1, CDK2, CDK4have actually been in decline (p<0.05), and with the increase of the concentration gradient, the decline was the more obvious, the protein expression of P27Kip1has risen significantly, and with the increase of the concentration gradient, the increase was the more obvious (p<0.05).(3) After Western blot method test, compared with the control, Methylprednisolone role in human glomerular mesangial cells, the protein expression of CyclinE, CyclinDl have actually been in decline (p<0.05), and with the increase of the concentration gradient, the decline was the more obvious. The protein expression of P27Kipl has risen significantly(p<0.05), and with the increase of the concentration gradient, the increase was the more obvious (p<0.05).Conclusion Methylprednisolone effect on human glomerular mesangial cells in vitro can significantly inhibit cell proliferation, and can make the protein expression of CyclinE, CyclinDl, CDK2, CDK4in the cells decreased significantly, make the protein expression of P27Kipl in the cells increased significantly, the down role and the rising role is related to the concentration. |
