Objective To investigate whether-238A/G and-308A/G polymorphisms in promoter region of TNF-a are associated with susceptibility to gout in Chinese Han male population.Methods238gout patients and263gout-free controls were enrolled. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect the genotype and allele frequencies of TNF-a-238A/G and-308A/G.Results No statistical difference was found in the genotype frequencies of TNF-a-238A/G and-308A/G polymorphism between the patients and controls. For TNF-a-238A/G:The genotype frequencies of AA, GG, and AG were0%,90.3%and9.7%respectively in patients and0%,93.5%and6.5%respectively in controls (χ2=1.741, DF=1, P=0.187, OR=0.646,95%CI=0.336-1.241). For TNF-a-308A/G:0%,86.1%and13.9%respectively in patients and0.8%,86.3%and12.9%respectively in controls (x2=1.893, DF=2, P=0.388). Neither did the allele frequencies of A and G in the two groups showed statistical difference. For TNF-α-238A/G:The allele frequencies of A and G were4.8%,95.2%in patients and3.2%and96.8%respectively in controls (x2=1.669, DF=1, P=0.196, OR=1.520,95%CI=0.802-2.882). For TNF-a-308A/G:6.9%,93.1%in patients and7.2%,92.8%in controls (x2=0.032, DF=1, P=0.857, OR=0.957,95%CI=0.590-1.552).Conclusion We can’t draw the conclusion that-238A/G and-308A/G polymorphisms in promoter region of TNF-a are associated with susceptibility to gout thus do not play a major role in the development of gout in Chinese Han male population from the present study. |
