Liver fibrosis (hepatic fibrosis) is the pathological process of a variety of liver injury factors caused by intrahepatic diffuse extracellular matrix (extracellular matrix,ECM) components excessive accumulation^the early stage of cirrhosis of the liver.Chronic hepatitis,liver fibrosis, cirrhosis, a serious threat to human health and life, has become a hot research of the medical profession at home and abroad, with the development of a better understanding of liver fibrosis, blocking of the early stage of liver fibrosis,mitigation and reversal of liver fibrosis process, is a effective treatment to prevent, delay cirrhosis, the recognization above has been gradually adopted by the pharmaceutical industry.But now chemical drugs has no effective and safe treatment for liver fibrosis drugs, Ttraditional Chinese Dugs with unique advantages in the field of liver fibrosis.Ultrafine grinding technology developed in recent years, is a interdisciplinary, cross-sector high-tech, but also a new developments and new applications of the old crush.It can improve the rate of cell breaking, increase the dissolution and bioavailability of the active ingredient of Chinese Medicines after ultrafine grinding, thereby enhancing clinical efficacy. In addition, it can save herbs, making a great practical significance to reduce the waste of the traditional Chinese medicine resources.In this paper, the Traditional Chinese Medicine Shierweichuanjia Tablet (later referred to as SWP), composed of Manis, Trioncis Carapax, Fructus Schisandrae, Raboniae and other traditional Chinese medicine was prepared, for cirrhosis with blood stasis, deficiency of blood loss, water poisoning caused by perverse trend not eliminate, applied to cirrhosis quiescent or activities period, with a good efficacy of anti-human liver fibrosis and early cirrhosis.In this paper, the micronization technology was adopted to grind on Fructus Schisandrae (later referred to as F.S),the particle size and pharmacokinetic in vivo of different the granularity powder was studied, and dissolution behavior in vitro of the SWP prepared by F.S of different granularity were investigated,providing an experimental basis for particle size choice of F.S.The results show that small the micropowder has a smaller particle size and narrow distribution;The the speed and extent of dissolution of the Deoxyschizandrin (later referred to as DS) in micropowder is higher than ordinary powder; rat plasma concentration curve fitted by DAS2.0program, the ultrafine powder of DS in the AUC(0-8) was58.853and the2.71times that of ordinary powder.In this paper, serum pharmacology was applied, observation of growth of HSC-T6cells in vitro affected by fine-powder or micropowder of the Trioncis Carapax. Using multi-factor orthogonal experiments, the single factor inspections, optimization the methods of preparation serum containing drugs, and determine the serum amount added into the system, MTT method was used to assay the inhibition on HSC-T6affected by the serum.The results showed that the water decoction of ultrafine has a strong inhibition of HSC-T6proliferation compared with fine-powder.Therefore,we choose the ultrafine crush on the Trioncis Carapax.In this paper,by investigated on micronization process of F.S and extraction process of Paeonia and other nine herbs, and ultimately determine the best preparation process of the SWP was:Smashed the Manis Souama and Trioncis Carapax into ultrafine powder, filtered; micronized F.S; Paeonia lactiflora Pall and other nine herbs,plused water8times the amount of the all drugs and reflowing2times, each time one hour,combined the extracts,filtered,the filtrates was concentrated to a relative density of1.10-1.12of the extract;taking the powder above added into the concretes, mixing, granulating,dried at70℃, added into3%carboxymethyl starchsodium and0.3%magnesium stearate,stired evenly, pressing tablets,and filming coat, that too.In this paper,quality standards and stability of the SWP has studied and a strict quality standard was formulated, the indicators detectded within the prescribed limits; accelerated test were observated the sample placed6months and long-term test were12months,all indicators was consistent with the0month.In this paper,with pharmacodynamic studies of the anti-liver fibrosis induced by carbon tetrachloride in rats of SWP,observing alanine aminotransferase (ALT), aspartate aminotransferase (AST) as other as some other biochemical indices and liver tissue hydroxyproline (HYP), collagen content in the rats,and liver histopathology were observed pathological changes.The results showed that the SWP can significantly reduce the serum ALT and AST activity,and improved the liver tissue degeneration obviously.In this paper, the acute toxicity on mice of the SWP was studied.Results have showed:the two weeks after intragastric administration in mice without a mouse died, and no significant toxicity; the largest quantity of administration of mice was60g kg-1,600times the adult daily dosage.In summary, the traditional Chinese medicine Shierweichuanjia Tablet has a good efficacy of anti-liver fibrosis and early cirrhosis.This preparation will produce good economic and social benefits, has a broad market prospect.
