Objective Study on the material basis of Radix Astragali in bufeihuoxue capsule with pharmacy and pharmacokinetics point of view. Optimize Radix Astragali extraction and purification process with the main component elements of matter as an index. Establish the draft quality standards of Radix Astragali extract. Lay the foundation for the development of bufeihuoxue capsule.Method In this study, high performance liquid chromatography and ultraviolet spectrophotometry was used to analysis the major components contained in the extract of Radix Astragali. The material basis of Radix Astragali was initially identified with in vivo intestinal perfusion model, the plasmatic and urinary constituents of Radix Astragali analysis. The material basis was considered as the index, used the orthogonal experimental design to optimization of extraction process of Radix Astragali. Determine the enrichment of Radix Astragali extract purification. Development of quality standards extract according to the general requirements for new drug development.Result The flavonoids of Radix Astragali’s separation was achieved on a AgilentTC-C18 column (250×4.6mm,5μm), monitored by UV absorption at 203nm and operated at 1.0 mL·min-1 using the gradient elution of Acetonitrile and water.Determination of Saponins in the extract of Radix Astragali was performed by UV spectrophotometry monitored by UV absorption at 544nm. Intestinal perfusion experiments, Radix Astragali extract eight kinds of main flavonoids components and saponins are absorbed to varying degrees.into the blood constituents in poor reproducibility of the three results are obtained with the number of component peaks were 8,3,3; into the urinary constituents, target constituents by the number of peaks for 4.Confirmed the initial structure, two of the eight kinds of flavonoids are calycosin and formononetin.The eight kinds of main flavonoids components and saponins are the Radix Astragali in bufeihuoxue capsule play in the material basis for efficacy. Calycosin, formononetin and saponins are considered as the index to optimizate the process of extraction and purification. Optimal extraction process of Radix Astragali:Radix Astragali followed by 12 times,9 times the amount of 80% of ethanol twice, each 1 hour. Purification showed that:the D101 macroporous resin as a filler, the liquid concentration of extract was 0.6g Radix Astragali·mL-1, the sample was 1BV, adsorption flow rate of 1 mL·min-1.2h after adsorption, desorption stage with 5 BV water,5BV20%ethanol and 6BV80% ethanol.The 80% ethanol was part of the collection, ethanol recycled and powder received. Quality study focuses on character, identification, determination of target components, inspection and other aspects of research, developed the draft quality standards of psoralen extract.Conclusion Initially clear the material basis of Radix Astragali in bufeihuoxue capsule. Optimize its extraction and purification process. Develop the Radix Astragali extract quality standards.Lay the foundation for the further development of bufeihuoxue capsule.
