Our country has thehighest rates in incidence and mortality of esophageal cancer in the world. Themortality rate ranks the fourth in malignant tumor in our country, and highlyranks the second in northern of China, only inferior to the gastric cancer. Bycase type, Esophageal cancer can be divided into esophageal squamouscarcinoma and adenocarcinoma. In our country, the morbidity of squamouscarcinomas is much higher than adenocarcinoma. Tentative diagnosisconstantly to be middle-late stage. The rate is low in5-year survival aftersurgery, the early cancer, relatively, its rate can be as high as90%in the5-year survival after surgery. Therefore, to strengthen the research ofesophageal cancer pathogenesis has important significance for its earlydiagnosis and improvement of surgical recovery.By the modern molecular biology research, the occurrence anddevelopment of the tumor relates to the apoptosis of cancer cells and theinhibitor of tumor blood vessel growth and other factors. Its mechanism iscomplicated, involving multiple genes and proteins. ID was found in leukemiacells from mice found in a CDNA library by Robert Benezra team in1990.It isa member of HLH transcription factors. ID exists in most mammalian cellswith four seed body ID-1,ID-2,ID-3,ID-4.The study found that the HLHfamily influence transcription regulation function. Generally the role of IDgene families similar to oncogene. ID protein can promote cellproliferation, and inhibite cell differentiation inducing cell apoptosis andpromote tumor cell cycle regulation function such as tumor metastasis andtumor angiogenesis. The research results show that ID-1protein expression have the effect that make the cells immortalized. So ID gene is regarded as apotential cancer gene. Its mechanism is through the inhibition of transcriptionlevel in p16ink4a and p21WAF1. This, in turn, can contribute to CDK4combine with CDK2and pRb, make pRb phosphorylate, then E2F protein getrid of the shackles of pRb, promote more cancer gene transcription, againmake cells to enter S phase. The study of Promoting cell over proliferationshows that ID-3plays a role similar to oncogene in the inhibition of celldifferentiation, cell proliferation and apoptosis induction and promoting. Mostcell biological processes have ID-3genes involved. Therefore, ID-1and ID-3play an important role in the process in tumor occurrence anddevelopment.VEGF(vascular endothelial growthelial growth factor) is amember of the dimer glycoprotein growth factors, promoting the proliferationof endothelial factor. It plays an important role in tumor induced newhorizontal vessels, increased vascular permeability, etc. VEGF is the moreimportant and specific cytokines in the found adjustment factors. It iscytokines to promote the growth of vascular endothelial cell. It can not onlypromote endothelial cell proliferation, but can promote vascular leakage, so itplays an important role in the formation of blood vessels. To make throughincreased vascular tumor cells to obtain large amounts of oxygen machine, and then make the tumor cell proliferation. At present, the research about threein esophageal squamous cell carcinoma is less at home and abroad. This studydiscusses the relationship between ID-1, ID-3, VEGF and occurrence anddevelopment of esophageal cancer (ESCC) which detected in40thoracicsurgery resection cases of esophageal squamous cell carcinoma (ESCC) in thefourth hospital, hebei medical university in July2012-in November2012, bywestern blot method. Methods: The expression of ESCC was detected in40thoracic surgeryresection cases of esophageal squamous cell carcinoma (ESCC) in the fourthhospital, hebei medical university in July2012-in November2012.Postoperative pathology were confirmed by diagnosis of pathology, in thefourth hospital, hebei medical university. Take10cases tissue adjacent to carcinoma (surgery to remove cancerous parts more than5cm distance fromtissue) as normal controls. According to their age, gender, differentiationdegree pathological type of tumor, pathological type Tumor infiltratingextent, Presence of lymph node metastasis and so on, it makes Groupingcomparison, respectively. It tests the expression of ID-3and VEGF insamples by western biot method.Statistical methodsResults:1ID-3positive expression in esophageal cancer tissues0.8255±0.10982,in adjacent tissues, the positive expression was0.1910±0.00876the P tissuetypes=0.000, the difference between the two was statistically significant (P<0.05).2In various degrees of differentiation esophageal ID-3positiveexpression were:0.7888±0.07570, moderately differentiated carcinoma, poorly differentiated carcinoma0.9986±0.07403, P pathologicalclassification=0.000, the difference between the statistics significance (P<0.05).3VEGF in esophageal cancer tissue positive expression rate was0.9438±0.10748, while the positive expression rate in the adjacent tissues0.4290±0.02378, P tissue types=0.000, the difference between the two wasstatistically significant (P <0.05).4in the presence of lymph node metastasis in esophageal squamous cellcarcinoma, VEGF in the positive expression rate respectively LNM (-):0.8461±0.06089,LNM (+):1.0236±0.05996, Plymph node metastasis=0.000,the difference was statistically significant (P <0.05).5pearson correlation analysis shows r=0.913, P=0.000, suggesting thatID-3and VEGF expression in esophageal squamous cell carcinoma werepositively correlated (P <0.01)Conclusion:1ID-3ESCC tissues was significantly higher than normal tissue, prompted Id-3expression may be related to the occurrence and ESCC. 2ID-3high expression may be related to the degree of malignancy, thelower and the higher the ID-3expression in ESCC tissue differentiation.3ESCC tissue VEGF expression was significantly higher than that ofnormal tissue, suggesting that VEGF expression may be related with ESCC.4VEGF expression may be associated with tumor invasion andmetastasis, the higher the expression of VEGF in ESCC lymph nodes, theeasier transfer.5ID-3and VEGF were positively correlated, suggesting that the presenceof ID-3activation and the activation of VEGF interaction, and jointly promotethe development of tumors in ESCC. | | Keywords/Search Tags: | esophageal squamous cell carcinoma, ID-3, VEGF, invasion, transfer, western blot | PDF Full Text Request | Related items |
