| Objective:To explore the expression of phospholipase Cγ-2(PLCγ-2),lipoxygenase-12(12-LOX), arachidonic acid (AA) in laryngeal squamous cellcarcinoma and para-carcinoma tissues and to study the the relationshipbetween lipid metablism and clinic stage and histological differentiation ofcarcinoma. Further, we investigate the role of lipid metabolism in theoccurrence and development of the laryngeal squamous cell carcinoma inorder to provide molecular biology evidence for the clinical strategy design.Methods:30cases were chosen from the Second Hospital of HebeiMedical University from2011/2to2012/5which clinical data were completeand were confirmed by pathological examination before operation. All of thepatients were male, whose age was from53to70and the average age was60.15.3years old. The patients had received neither radiotherapy norchemotherapy before the operation who had not hyperthyroidism orhypothyroidism. We took the laryngeal squamous cell carcinoma andpara-carcinoma tissues as the objects of the research. The para-carcinomatissues were gained from the edge of the tumor which distance exceeded0.5cm (confirmed to be normal laryngeal tissues by pathology). According to theInternational Anti-Cancer Coalition (UICC)2002standard, the cases wereclassified by its clinical features. Of the30cases,18were in stage I~II,12were in stage III~IV. According to the tumor differentiation degree,19caseswere well differentiated,11cases were moderately and poorly differentiated.14cases were with lymph node metastasis and16cases were not which wereconfirmed by pathology, According to the age,19cases were in53-60-year-old group, and11cases were in61-70-year-old group. The expression ofPLCγ-2and12-LOX in carcinoma tissue and para-carcinoma tissues was detected by Streptavidin-perosidase method (SP method) and the content ofthe arachidonic acid in carcinoma tissue and para-carcinoma tissues wasdetected by gas chromatography/mass spectrometry(GC/MS). Some part ofspecimen was put into the10%formalin fixative immediately after surgery,conventional paraffin-embeded, sectioned by5μm thick slice, deparaffinagedby xylene in convention, dehydrated by alcohol in gradient, removedendogenous peroxidase, antigen repaired by microwave. The expression ofPLCγ-2and12-LOX was detected by immunohistochemical method. Then theslice were stained with DAB, hematoxylin stain, dehydrated, transparentedand mounted by gum. The other part of the specimen were put into normalsaline and were prepared for gas chromatographyï¼mass spectrometry. Allexperimental data were analyzed by SPSS13.0statistical software whichP<0.05is statistical significance.Results: The positive expression rate of PLCγ-2in carcinoma tissue wassignificantly higher than it in para-carcinoma tissues and the difference wasstatistical significance (P <0.05). The positive expression rate of12-LOX wassignificantly higher in carcinoma tissue than it in para-carcinoma tissues andthe difference was statistical significance (P <0.05). The content ofarachidonic acid was significantly lower in carcinoma tissue than it inperitumoral normal laryngeal mucosa tissues and the difference was statisticalsignificance (P <0.05).The positive expression of PLCγ-2was closely related to the clinic stageand histological differentiation of larygeal squamous cell carcinoma. Itspositive expression in I~II stage was lower than that in III~Ⅳstage. Thepositive expression of PLCγ-2in well differentiated, moderately differentiatedand poorly differentiated laryngeal squamous cell carcinoma was significantdifference (P <0.05), which showed adscendent trendency.The positive expression of12-LOX was immediately related with clinicstage and histological differentiation of carcinoma. Its positive expression inI~II stage was lower than that in III~Ⅳ stage. The positive expression of12-LOX in well differentiated, moderately differentiated and poorly different- iated laryngeal squamous cell carcinoma was significant difference (P <0.05)which showed adscendent trendency.The content of arachidonic acid was immediately related with clinic stageand histological differentiation of carcinoma. Its contnet in I~II stage waslower than that in III~Ⅳ stage.Its content in well differentiated, moderatelydifferentiated and poorly differentiated laryngeal squamous cell carcinomawas significant difference (P <0.05),which showed adscendent trendency.The positive expression of PLCγ-2,12-LOX and the content ofarachidonic acid were ralted to the metastasis of laryngeal squamous cellcarcinoma. Their positive expression in lymphatic metastasis group washigher than that in no lymphatic metastasis group and the difference wassignificant (P <0.05),so was the content of arachidonic acid.The expression of PLCγ-2and12-LOX had no significant correlationwith age (P>0.05) and the content of arachidonic acid had no significantcorrelation with age either.(P>0.05).Conclusions: Overexpression of PLCγ-2,12-LOX and the content ofarachidonic acid were closely related with clinic stage, pathological gradingand lymphatic metastasis but not related with age. With the development ofcarcinoma and its lymphatic metastasis, the expression of PLCγ-2,12-LOXand the content arachidonic acid were higher. The lipid metabolism disorderinduced by PLCγ-2, AA and12-LOX plays an critical role in tumor genesis,invasion, and metastasis of the laryngeal carcinoma and these gene could beused as endogenous molecular markers in tumor prognosis.... |
