Objective:According to the molecular epidemiological investigation of dental caries from Yugur and Han population in Gansu area, in order to investigate the relationship between the distribution of genotype and allele frequencies in the three single nucleotide polymorphisms (SNPs) of carbonic anhydrase-VI gene (CA-VI) and the severity of dental caries.Material and Methods:Choose a total of355subjects in Yugur autonomous counties of Gansu province and conduct a high caries group-low caries group for comparison study in November of2011. DNA was extracted from2ml peripheral venous blood using genomic DNA extraction kit. SNPs in the CA-VI gene were analyzed using multiple PCR and MassArray, the genotype and allele frequencies of rs2274328(A/C)、rs17032907(C/T) and rs1157666(A/C) were detected by them directly. The differences between the high caries group and low caries group were analyzed by chi square test, and the data analysis was used by Haploview4.2and SPSS software package.Results:1. All genotypes and alleles in the three SNPs of rs2274328(A/C)、rs17032907(C/T) and rs1157666(A/C) of CA-VI gene were in Hardy-Weinberg equilibrium(P>0.05).2. A linkage disequilibrium was noted between three SNPs of rs2274328(A/C)、 rs17032907(C/T)and rs1157666(A/C)within CA-VI gene(D’>0.9). It demonstrated that the wild haplotype (ACA) might be associated with low DMFT index (P=0.015, OR [95%CI]=0.635[0.440-0.918]).3. We found that the frequency of the rs17032907TT genotype was significantly increased in the high caries group compared with low caries group in the whole population (P=0.001), and TT genotype increased the risk of high caries (OR [95%CI]=2.144[1.096-4.195]).4. After ethnic stratification, rs2274328CC genotype was related to the occurrence of high caries in Yugur population (P=0.014, OR [95%CI]=3.5[1.15010.649]).5. There were no statistically significant differences between the frequencies of alleles of rs2274328(A/C)、rs17032907(C/T) and rs1157666(A/C) within CA-VI gene(P>0.05).Conclusion:Our results suggest that the rs2274328(A/C)、rs17032907(C/T) and rs1157666(A/C) polymorphisms could affect the susceptibility of dental caries in Gansu area, and maybe served as a test indicator for the vulnerable groups with dental caries. In addition, the polymorphism of rs2274328in the gene of CA-VI was probably different in different population and regions. However, further studies with larger numbers of samples of different ethnic groups are required for further evaluation to confirm our findings.
