| Objective:in recent years, numerous studies show that tumor occurrence, development and cell cycle control disorders, DPC4/SMAD4gene is the most important tumor suppressor genes in the cell cycle control system and its loss of expression for tumor formation play a key role. Involved in TGF-β signaling channels regulate the cell cycle and signal transduction. This article by real-time quantitative PCR method to detect the expression levels the SMAD4mRNA in patients with colorectal cancer, the relationship between the analysis with clinicopathological parameters of patients with colorectal cancer. Further demonstrate the role of DPC4/SMAD4gene in colorectal tumors and pathological correlation in colorectal tumors, this paper aims to explore the the SMAD4ability to become important biological markers of colorectal tumors, thereby increasing the rate of clinical diagnosis for colorectal tumors early diagnosis, early treatment, prognosis and future targets of colorectal cancer gene therapy provide a certain basis.Methods:In this study, the real-time quantitative RT-PCR was used to detect extracted with pathologically confirmed40cases of colorectal cancer patients and20healthy human peripheral blood DPC4/SMAD4mRNA expression, and reference gene GAPDH, using2-the△△ct, analysis,2-△"represents a multiple of the expression level of target gene expression in cancer tissue relative to normal tissue. Smad4expression (1) in patients with colorectal cancer was significantly lower than the normal group (P<0.05), and (2) of Smad4expression with age, gender, tumor site unrelated (P>0.05), but the Dukes staging of colorectal tumors lymph node metastasis, histological grade related(P<0.05).Conclusions:Real-time quantitative RT-PCR detection of colorectal cancer patients with sensitivity, specificity SMAD4mRNA expression in peripheral blood, aid in the diagnosis of colorectal cancer patients, determine treatment, to determine the efficacy, whether metastasis and prognosis has important clinical value. Low SMAD4gene expression in colorectal cancer patients with poor prognosis, the expression levels and clinicopathological staging of colorectal cancer, lymph node metastasis was inversely related; DPC4/SMAD4mRNA expression levels and colorectal cancer prognosis and clinical staging, lymph node metastasis, but may not be independent prognostic factors necessary bulk experiments, better follow-up and more state-of-the-art experimental techniques further validation. |
