The Daqinglong Decoction Antipyretic Drug Substance And Serum Pharmaceutical Chemical Research

Daqinglong decoction, which comes from Shanghan Lun written by ZhangZhongjing, is made up of Ephedrae herba, Cinnamomi Ramulus, ArmeniacaeSmen Mrum, Gycyrrhizae Rdix Et Rhizoma Praeparata Cum Melle, JujubaeFructus, Zingiberis Rhizoma Recens and Gypsum Fibrosum.The decoction is aclassic prescription with the exact clinical efficacy and has been proved bypractice through thousands of years. Clinical studies show that, it could treatcolds, fever, no sweat disease, chronic bronchitis, asthma and so on. Its curativeeffect is exact. And modern pharmacodynamic studies indicate that theprescription has antipyretic, antibacterial, antiviral, enhance the phagocytosisfunction of macrophage. So far, research of daqinglong Decoction mainly focuson clinical application, its efficacy material basis of rare reports.This dissertation considered the decoction as a whole. The herb wasextracted and selected by supercritical fluid extraction and systematic solution.The extracts were as following: extracts by SFE-CO2，alkaloids，phenolicacids，glycosides，total polysaccharide and gypsum Decoction.Through thescreening of the fever induced by lipopolysaccharide test rats, to determine theeffective parts of daqinglong decoction antipyretic material of alkaloids and totalpolysaccharide.Analysis by high performance capillary electrophoresis（HPCE）, the reference fingerprint on the requirements of the state food and drugadministration (SFDA)" fingerprint technology requirements of traditionalChinese medicine injection (Provisional)", HPCE fingerprint map of daqinglongdecoction alkaloid part was established, calibrated. We determined13peaks ofephedrine and pseudoephedrine were assigned. Using GC-MS, we analyzed theextracts of total polysaccharide in daqinglong decoction, and identified8kinds ofchemical compounds. Analysis results show that, the structure of polysaccharidesis composed of L-Arabia sugar, D-mannose, D-galactose, D-galacturonic acid,D-glucose, L-rhamnose, D-xylose and D-ribose and so on.We are respectively make studies on serum pharmacochemistry of alkaloidsand polysaccharide part of daqinglong decoction. By using the method of HPCEanalysis, fingerprint of alkaloid constituents absorbed into blood from theidentified main effective components of ephedrine and pseudoephedrine. UsingGC-MS analysis, blood from polysaccharide constituents identified glucosederivatives.This dissertation use interdisciplinary research methods to preliminaryinterpretation of the material basis of daqinglong decoction antipyretic, providesthe scientific basis for the clinical effect of the classic formula, and for thedevelopment of new varieties of Chinese medicine hospital preparations and lay acertain foundation.
