Recently years, type2diabetes has become a hazard to our country andeven the world human health. Worldwide, about170million patients,including type2diabetes (T2DM) for more than90%.At present, China hasalready over taken India as the first big global diabetes. and, The groups ofdiabetes high-risk are expanding in China, about150million. Although after atimely and effective treatment which can control blood sugar strictly, delayand/or prevent the occurrences of diabetes complications, improve patient’ssurvival rate. But this after brings economic and mental pressure to all patients.So, seeking effective and reasonable prevention measures to reduce theincidence of diabetes is of great significance.Different species of animals T2DM model are proved to have differentlevels of insulin rise in the early. But for insulin level’s long-term research isless. This experimental preliminary study on this question.Objective: To observe the changes of insulin level of T2DM rat model.Methods:80healthy Sprague Dawley rats were randomly divided into4groups:Ⅰ High sugar and fat F: feed with high sugar and high fat.Ⅱ High sugar and fat+Streptozocin:feed with high sugar and high fat,after six weeks,Intraperitoneal injection of Streptozocin(30mg/Kg).Ⅲ Ordinary feed: Ordinary feed.Ⅳ Ordinary feed+Streptozocin:Ordinary feed,after six weeks,Intraperitoneal injection of Streptozocin(30mg/Kg).This experiment adopts the high sugar and high fat feed SD rats.after sixweeks,intraperitoneal injection of STZ30mg/Kg,after72hours,blood sugartendency is greater than or equal to11.1L, and preparation for7days,whichMeans that the model is success. Tested blood sugar by Yi Cheng blood glucose meter, tested serum insulin levels by using rat insulin ELISA, testedmRNA expression levels in pancreatic beta cells by RT-PCR. After thesuccess of the model,before injected STZ,in the3days,4weeks,8weeks,12weeks,16weeks,18weeks,28weeks, tested the serum insulin levels andmRNA expression levels in pancreatic beta cells.Results:1glucoseIn0days, the glucose of groupⅠ and Ⅱ is higher than group Ⅲ and Ⅳ,but there was no significant difference between groups (P>0.05); In groupⅡ, glucose has a significant risein the3day, compared with the zero therewas statistically significant (P=0.001<0.05), but no difference at each timepoint after; In group Ⅰa nd Ⅲ,every time has no statistical difference;(P>0.05); In group Ⅳ,glucose has a significant rise in the3day, comparedwith the zero there was statistically significant (P=0.001<0.05),And after the4week each time were relatively low, compared with the0day there is nostatistical difference (P>0.05).2Serum insulin levels (ELISA)In groupⅠ,serum insulin level were no statistical difference betweendifferent time points (P>0.05); Compared with group Ⅱ, at the same time,itsinsulin level is Lower, but had no statistical difference (P>0.05);Comparedwith group Ⅲ, the0day is higher, there is statistical difference(P=0.001<0.05), The rest of the points at the same time insulin level is high,but had no statistical difference (P>0.05);Compared with group Ⅳ,the0dayis higher, there is statistical difference (P=0.044<0.05), The rest of the pointsat the same time insulin level is high, but had no statistical difference(P>0.05).In group Ⅱ,serum insulin level begins to rise in the3day(30.393±2.038), but compared with the0day there is no statistical difference (P>0.05),in the4week(31.321±2.467) it increased significantly, compared with the0day、3day,difference existed(P<0.05), in the4week、8week、12week、16week、18week, serum insulin was at a higher level, but there was no significant difference (P>0.05), but showed a trend of falling after rising first,highest when the12week(37.071±5.537). At the28week(30.854±3.420) ofsignificantly lower serum insulin levels, compared with the4week、8week、12week、16week、18week,difference existed(P<0.05), but compared withthe0day、the3day have no difference (P>0.05); Compared with group Ⅲ,the0day is higher, there is statistical difference (P=0.001<0.05), The rest ofthe points at the same time insulin level is high, but had no statisticaldifference (P>0.05). Compared with group Ⅳ, the0day is higher, there isstatistical difference (P=0.024<0.05), The rest of the points at the same timeinsulin level is high, but had no statistical difference (P>0.05).In group Ⅲthe level of the serum insulin was no significant alteration,various periods were no statistical difference (P>0.05); Compared with groupⅣ, at the same time,insulin level is high, but had no statistical difference(P>0.05).In group Ⅳ,serum insulin level presents the downward trend all over thetime, but no statistical difference between the time points (P>0.05).3The expression level of mRNA of pancreatic beta cellsIn groupⅠ,The expression level of mRNA of pancreatic beta cells wereno statistical difference between different time points (P>0.05); Comparedwith group Ⅱ, at the same time,its expression level of mRNA of pancreaticbeta cells is Lower, but had no statistical difference (P>0.05);Compared withgroup Ⅲ,the0day is higher, there is statistical difference (P=0.001<0.05),The rest of the points at the same time expression level of mRNA ofpancreatic beta cells is high, but had no statistical difference(P>0.05);Compared with group Ⅳ,the0day is higher, there is statisticaldifference (P=0.044<0.05), The rest of the points at the same time expressionlevel of mRNA of pancreatic beta cells is high, but had no statisticaldifference (P>0.05).In group Ⅱ,the expression level of mRNA of pancreatic beta cellsbegins to rise in the3day, but compared with the0day there is no statisticaldifference (P>0.05), in the4week it increased significantly, compared with the0day、3day,difference existed(P<0.05), in the4week、8week、12week、16week、18week, expression level of mRNA of pancreatic beta cells was at ahigher level, but there was no significant difference (P>0.05), but showed atrend of falling after rising first, highest when the12week.
